Identification of synonyms and homonyms in grapevine cultivars (Vitis viniferaL.) from Asturias (Spain)

Abstract: SUMMARYThe abandonment of Asturian vineyards over the last century almost resulted in the extinction of the grapevine crop and major cultivar confusion. Due to the restoration of a number of these vineyards in recent years, research into the regional varietal stock is needed. The aims of the present study were to characterise genetic resources of Vitis vinifera L. in Asturias and to identify synonyms and homonyms through an analysis of six microsatellite markers: VVS2, VVMD5, VVMD7, VVMD27, VrZAG62, and VrZAG7… Show more

“…The PCR reaction was checked in 3% agarose gels and PCR products were analyzed on an ABI PRISM ® 3100 Genetic Analyzer (Applied Biosystems) as described in Moreno-Sanz et al (2008). The internal standard used to assign sizes to the DNA fragments was GENESCAN500LIZ.…”

“…The cultivars endorsed by the regulations of this appellation are used in new vineyards, as well as in the replanting of old ones, leading to the extinction of other cultivars. The loss of grapevine phytogenetic resources in the Asturian vineyard is becoming obvious, as no stock of some cultivars (Carrascón, Agudiello, Rondal Negro, Mallén, Pardusco Prieto, Picudo, Rondales, Conrasión and Bondal), mentioned in bibliography as existing before the phylloxera attack (García, 1914;Naredo, 1914), have been reported in recent studies of ampelographic and genetic characterization Pérez, 1999, 2000;Martínez et al, 2002;Moreno-Sanz et al, 2008;Santiago et al, 2005). The aim of the present study is to analyse Asturian grapevine diversity through microsatellite markers (SSRs).…”

“…It provides fruity, delicate wines which are dark cherry in colour. Verdejo tinto is a synonym of the Merenzao cultivar from Galicia (Moreno-Sanz et al 2008); it produces rosé wines with a low anthocyanins concentration. There exist four clones of this cultivar that have recently been certified in Spain, three of which have not yet been commercialized.…”

“…DNA extraction and amplification was carried out as in Moreno-Sanz et al (2008). PCR products were analysed by means of an automated ABI PRISM  3100 DNA sequencer (Applied Biosystems, Foster City, USA) and the fluorescence labelled fragments were sized using the Genemapper Software v.4.0 (Applied Biosystems).…”
