Identification of targets of monoclonal antibodies that inhibit adhesion and growth in Mycoplasma mycoides subspecies mycoides

Abstract: HighlightsA panel of anti-Mmm mAbs was produced and screened for host-pathogen inhibition.13 mAbs inhibited adhesion of Mmm to host target cells.Anti-capsular polysaccharide inhibited growth and caused agglutination of Mmm.Anti-PDHC inhibited adherence of Mmm cells showing the possible role of glycolytic enzymes in host-pathogen interaction. One novel antigen that is a promising vaccine candidate against CBPP identified.

“…Also, polyclonal rabbit serum against recombinant MSC_0267 blocked the adhesion of Mmm to BoLEC by 41%. Further in vivo studies are required for exploring the immune response induced by Mmm antigens recognized by these antibodies (Aye et al, 2018). A precision-cut lung slices (PCLS) infection model for Mmm has been established to study host-pathogen interactions.…”

“…Mathematical models have been developed to evaluate economic impact, transmission dynamics, and the potential impact of various control strategies [256,274,[279][280][281][282][283]. In vitro models utilizing bovine lung epithelial cells and a variety of assays have been described [240,284,285]. Bovine respiratory explants from trachea, bronchi and lungs of slaughtered cattle are a promising ex vivo tool for further investigation of CBPP infection [286].…”

“…120 However, current data indicate that the adhesion of respiratory mycoplasmas is a more complex and multistep process, which reasonably involve both specific (ie, ligandreceptor interactions) and nonspecific (hydrophobic interactions) mechanisms. 8 Mycoplasmas have long been considered extracellular parasites, strictly adhering to the host cell membrane to acquire nutrients. However, this dogma has been largely disavowed, thanks to the development of suitable in vitro models of study and more sophisticated microscopic technologies.…”

“…Recent evidence indicates that Mmm adhesion molecules are immunogenic, as the organism attachment is inhibited by immune sera. 8,28 Unlike other mycoplasmas, the capsular galactan (a polysaccharide made of 6-O-b-D-galactofuranosyl-D-galactose) seems to reduce the attachment of Mmm to host cells, probably masking the adhesion molecules that become less exposed to the immune system. 93 A protein ("P19") has also been suggested to contribute to Mmm adhesion in vitro.…”
