Identification of Two Human β-Tubulin Isotypes

Abstract: The sequence of a human beta-tubulin cDNA clone (D beta-1) is described; our data revealed 95.6% homology compared with the sequence of a human beta-tubulin processed pseudogene derived by reverse transcription of a processed mRNA (Wilde et al., Nature [London] 297:83-84, 1982). However, the amino acid sequence encoded by this cDNA showed less homology with pig and chicken beta-tubulin sequences than the latter did to each other, with major divergence within the 15 carboxy-terminal amino acids. On the other ha… Show more

“…The pRI clone was kindly provided by S. McKnight. The same filter was washed free of probe and rehybridized with a 0.8-kilobase (kb) PstI fragment of a cDNA human ,-tubulin cDNA clone (14). (C. Whitfield provided the 1-tubulin clone, which was isolated in the laboratory of N.…”

“…This initial alignment was then filtered for both duplicate and fragmentary sequences, producing a final set of 23 unique sequences. Based upon differences found in each protein's carboxyterminal tail region, ten consensus sequences corresponding to tubulin isotypes βI, βIIa/b, βIII, βIVa/b, βV, βVI, βVII and βVIII, were characterized (figure 1) [36][37][38][39][40]. Of these Sequence alignment of β tubulin isotypes.…”
