Identification of Two Species of Yeast-like Symbiotes in the Brown Planthopper, Nilaparvata lugens

Abstract: To determine the species of the yeast-like symbionts (YLS) in the brown planthoppers (BPH), Nilaparvata lugens, YLS were first isolated and purified by ultracentrifugation from the fat bodies of BPH, and then 18S rDNA and internal transcribed spacer (ITS)-5.8S rDNA sequences of YLS were amplified with the different general primers for fungi. The results showed that the two different 18S and ITS-5.8S rDNA sequences of YLS were obtained. One 2291-bp DNA sequence, which contained 18S and ITS-5.8S rDNA, showed the… Show more

“…The anatomical organization of bacteriocytes varies, including a single aggregation of cells in the body cavity (e.g., in aphids and whiteflies) or paired structures associated with the lateral wall of the abdomen (in leafhoppers and cicadas) (Buchner 1965). Alternative structural arrangements include the localization of bacteria to the distal midgut of plataspid stinkbugs and of yeasts to the fat body of some planthoppers (Dong et al 2011; Nikoh et al 2011). These microorganisms are vertically transmitted, usually via the female ovaries, such that the cytoplasm of each egg bears a maternal inoculum of symbionts at oviposition; gut symbionts are deposited externally, and acquired by the feeding offspring (Buchner 1965).…”

“…Simbiosis ini membantu serangga mendapatkan nutrisi, membantu perkembangan, reproduksi, dan pertahanan terhadap musuh alami (Chen et al 2011;Dong et al 2011;Tang et al 2010). Endosimbion WBC terdiri atas jamur berfilamen atau khamir (yeast-like symbiotes, YLS) yang bersifat obligat dan bakteri obligat maupun fakultatif (Dong et al 2011;Fan et al 2015).…”
