“…In addition, freshly isolated cells identified to express high amounts of c-kit were analyzed by double immunofluorescence. In this procedure, cells were stained by antibodies against CD117, CD10 or CD34 which were then detected by PE-conjugated F(abЈ) 2 fragments. An excess of irrelevant mouse Ig was used to block free antigen binding sites on the anti-mouse Ig-PE.…”

“…This protooncogene encodes the specific surface receptor for the pluripotent hematopoietic colony-stimulating factor SCF. [1][2][3] SCF acts synergistically with other growth factors, including erythropoietin (EPO), G-CSF, macrophage (M)-CSF, GM-CSF, IL-3, IL-6, IL-11 and IL-12, [4][5][6][7] and plays an important role in normal and leukemic hematopoiesis. Mature lymphocytes exhibit little or no responsiveness to the c-kit ligand, SCF.…”

“…The in vitro properties of the recombinant rat and recombinant human SCF (rrSCF and rhSCF) suggest that SCF is a multipotent factor acting on cells of the myeloid, erythroid, mast, and lymphoid lineages [24- [26,271. No effect on human pre-B cell development has been detected for rhSCF and may be due to a limited role of rhIL-7 on early human B cell development.…”

“…This finding suggested that, based on the phenotypic similarities and complementary nature of the defects produced by the Wand Sl mutations, the ligand for c-kir was the product of the Sl locus [16, 191. Recent work from our laboratory and others has identified the ligand for c-kit and demonstrated that this ligand is a product of the Sl locus [20-261. In our own studies, we purified the natural form of this factor, which we termed Stem Cell Factor (SCF), from the supernatant of a buffalo rat liver cell line (BRL-3A) based upon the ability of SCF to stimulate primitive mouse hematopoietic progenitor cells [26]. Recombinant human and rat SCF have been produced in E. coli, and many of the biological properties have been described [23-261. …”

“…A renewed interest in these mutants came when the W and Sl gene products were characterized at the molecular level (Chabot et al, 1988;Geissler et al, 1988;Copeland et al, 1990;Huang et al, 1990;Zsebo et al, 1990;Williams et al, 1990). Thus, the complementarity of the two mutations was accounted for by the fact that the W gene encodes a transmembrane tyrosinekinase receptor, c-kit, the ligand of which turned out to be the product of the Sl gene.…”
