IL-1β(interleukin-1β) stimulates the production and release of multiple cytokines and chemokines by human preadipocytes

Abstract: The effect of IL-1β on cytokine and chemokine production by human preadipocytes has been examined. Preadipocytes were incubated with IL-1β, and cytokine and chemokine release was measured at 24 h by protein arrays, while the expression of cytokine/chemokine genes was assessed by qPCR at 4 and 24 h. IL-1β stimulated the secretion of multiple cytokines/chemokines, including IL-6, IL-8, IL-10, IL-13, MCP-4, TNFα and IP-10. IL-10 was not released by un-stimulated preadipocytes, while IL-6 exhibited the greatest re… Show more

“…38,39 It is well known in the literature that IL-1b increases IL-6 and IL-8 secretion. 14,[40][41][42] In a recent study, human preadipocytes were shown to be highly responsive to stimulation with IL-1b, resulting in release of various cytokines, including TNF-a, IL-6, IL-8, and IL-10. 40 Because protein production detection is paramount, results showed significant decrease in secretion of IL-6 and IL-8 induced by IL-1b in HGF and HPLF when AT1R was silenced.…”

“…These immunoassays provide a rapid, sensitive measurement of specific proteins in small sample volumes. Plates were pre-coated with antibodies on independent, discrete spots and the assay then performed essentially according to the manufacturer's instructions, as described previously (Alomar et al, 2015;Alomar et al, 2016). The data was analysed using Proprietary Meso Scale software; the lowest level of detection was 1.03 pg/ml for ICAM1 (interlot CV 17.8%), 6 pg/ml for VCAM1 (interlot CV 2.6%), 11 pg/ml for MMP1 and 2.1 pg/ml for MMP3 (mean intraplate CV <15% for both MMP1 and MMP3).…”

“…TNFα is well recognised to have a powerful stimulatory effect on the synthesis and secretion of multiple cytokines and chemokines in human adipocytes, as well as of MMPs (Wang et al, 2005;Cawthorn & Sethi, 2008). The effect of IL-1β on adipocyte function has, however, received much less attention than TNFα (Bing, 2015), but our recent studies demonstrate that this pro-inflammatory factor stimulates the expression of a number of cytokine and chemokines genes in human adipocytes and preadipocytes, and that the release of the encoded proteins is also stimulated (Alomar et al, 2015;Alomar et al, 2016). In the present study, we have examined the effect of IL-1β on the expression of multiple ECM and cell adhesion genes in human adipocytes using a pathway-focused PCR array.…”

“…Leptin, up-regulated by inflammatory mediators such as tumor necrosis factor (TNF), IL-1β, and insulin, promotes the release of proinflammatory cytokines such as TNF, IL-12, and IL-6 [9,10] . In obese individuals, increased leptin production leads to a reduction in adipose tissue-infiltrating regulatory T cells (T reg ), thus amplifying local inflammation [7,11] .…”
