IL-4Rα-expressing CD11c+ cells contribute to driving optimal cellular responses during Schistosoma mansoni infection in mice

Abstract: Development of IL‐4 receptor alpha (IL‐4Rα)‐dependent cellular immunity regulates host protection against acute schistosomiasis. In this study, we investigated the importance of IL‐4Rα‐expressing CD11c+ cells in driving the development of optimal cellular responses to Schistosoma mansoni infection by using CD11ccreIL‐4Rα−/lox BALB/c mice, which lacked IL‐4Rα expression on dendritic cells and alveolar macrophages. Abrogation of IL‐4Rα expression on CD11c+ cells affected activation of CD4+ T cells, resulting in … Show more

“…Namely, IL-4 receptor expression on alternatively activated macrophages and myeloid cells in general appears to drive specific aspects of S. mansoni pathology (44,45). IL-4 receptor is expressed by diverse cell types such as CD4 ϩ and CD4 -T cells (46,47), smooth muscle cells (48), B cells (49), FoxP3 ϩ regulatory T cells (50), and CD11c ϩ cells (51). This corpus of research suggested to us that urothelial IL-4 receptor could be important in the bladder pathogenesis of urogenital schistosomiasis.…”

“…Together with studies showing that the deletion of IL‐4Rα in DCs and monocytes impaired Th2 responses to the parasite Schistosoma mansoni [69], as well as the differentiation of Th2 cells and infiltration of eosinophils into the lung in a model of allergic airway inflammation induced by house dust mite [70], these observations suggest that IL‐13 skews DC function toward a Th2 stimulatory function, regardless of tissue location and of whether IL‐13 is produced at steady state or induced by cytokine signaling or antigen recognition.…”
