Psoriasis is an autoimmune disease that is characterized by the appearance of lesions on the skin. The current treatment aims to control the symptoms. The efficacy of Stachytarpheta jamaicensis (L.) Vahl’s for autoimmune rheumatoid arthritis and systemic lupus erythematosus has been tested in animal models. The aim of this research was to evaluate the effect of the water extract of S. jamaicensis leaves on psoriasis model animal (male Balb/c mice) induced topically by imiquimod. The water extract of S. jamaicensis leaves is made by boiling. The animal was divided into groups: normal, control, methotrexate 0.2 mg/kgBW, the extract (doses 25, 50 and 100 mg/kgBW). The measured parameters were the Psoriasis Area and Severity Index (PASI) score and skin histopathology. The results showed that all doses of the extract could reduce the PASI score when compared to the control group. Histological results showed that there was a decrease in keratin growth in test animals that were given the extract. Extracts at doses of 25 and 50 mg/kgBW can reduce the thickening of keratin in the epidermis of the back and ears. It can be concluded that the water extract of S. jamaicensis leaf has the most effective activity to prevent psoriasis recurrence in the dose range of 25 and 50 mg/kgBW.Keywords: Psoriasis, Stachytarpheta jamaicensis leaf water extract, PASI, keratin, imiquimod

Second-harmonic generation (SHG) microscopy is a label-free imaging method that can be used to visualize the detailed arrangement of collagen structures in biological tissues. Here, we sought to optimize the speed of microscopic SHG image acquisition of macroscopic fixed tissue sample areas by employing the wide-field imaging with a high power and medium, 1 MHz pulse repetition frequency laser in combination with a mechanical sample scanning. Unlike in the conventional laser-scanning microscopy, the optimum of the wide-field acquisition entails an interplay between the size of the illuminated area and the intensity of the generated signal. We delineate quantitative procedures to set the image parameters for the maximum speed of the tiled image acquisition, and also describe the possible optimization of the laser parameters for further enhancement of the speed of acquisition.

This research project is aimed to prepare improved topical preparation of Calcipotriol in the treatment of psoriasis by incorporation of an anti-inflammatory agent, keratolytic agent, humectants, surfactant, thickener & to perform its evaluation. In these different preparations of lotions sodium carboxymethylcellulose is used as a viscosity-enhancing agent in varying concentrations. This non-steroidal formulation is prepared by adding tea tree oil to avoid side effects associated with steroidal preparation and to avoid the issue of atrophy with long-term use of steroids. Evaluation of all the formulations such as spreadability, viscosity, drug content determination, pH, in-vitro release study, to choose the optimized formulation was carried out. According to ICH guidelines, stability studies were carried out to check the stability (parameters such as appearance, pH, viscosity, drug content) of prepared formulations for 3 months at room temperature. In-vitro drug release study CT5 formulation was carried by dialysis diffusion method. The cumulative% drug release of CT5 formulation in 210minutes was 85.10%. Based on the evaluation, formulation (CT5) was found to be better among all the formulations. The prepared calcipotriol formulation and a commercial formulation of calcipotriol were evaluated for anti-psoriatic activity. In-vivo anti-psoriatic was performed by using the Imiquimod Induced Psoriasis skin model. The results obtained in this study have been concluded that the prepared formulation (CT5) has great potential for topical delivery in the treatment of psoriasis.
