Immobilized metal ion affinity nanospheres for $\alpha $-amylase immobilization

Abstract: Immobilized metal chelate affinity chromatography (IMAC) support was practiced for α -amylase immobilization. Poly(hydroxyethylmethacrylate-methacryloylamidotryptophan)-Ni 2+ [p(HEMA-MAT)-Ni 2+ ] nanospheres, average diameter 100 nm, were produced by surfactant free emulsion polymerization. Characterizations of p(HEMA-MAT)-Ni 2+ nanospheres were carried out by Fourier transform infrared (FTIR) spectroscopy and scanning electron microscope (SEM). In addition, average particle size, size distribution, and surfac… Show more

“…α-amylase is one of the largest selling enzymes for a wide variety of industrial applications. They are endo acting glycoside hydrolases which hydrolyzes α-1,4-glycosidic bond present in polysaccharides to produce glucose, maltose and maltotriose units [25]. These low molecular weight products are widely applied in the food, paper, textile, baking and brewing industries [26].…”

“…Kalburcu et al [34] described that reusability of an enzyme immobilised on a matrix is a must for large-scale operations. Hence, a study on repeated uses of the immobilised glucoamylase on MCF silica (9.2T-3D) was conducted to know its activity after many saccharificarion processes as shown in Fig.…”

“…Thermal and storage stability were significantly superior for bound enzyme compared to the soluble form. Moreover, this support was repeatedly used for the immobilization of α-amylase [53]. Saware et al [54] described a novel approach to understand the interaction of amylase with gold (Au) and silver (Ag) NPs and their impact on its activity.…”
