Immune-enhancement effects of tuna cooking drip and its enzymatic hydrolysate in Balb/c mice

Abstract: Tuna cooking drip (TCD) is a protein rich by-product of canned tuna processing that is typically discarded. In this study, the immune-enhancing activities of TCD and its enzymatic hydrolysate (EH-TCD) were investigated by intraperitoneally administering Balb/c male mice with TCD and EH-TCD for 2 weeks. This administration resulted in an increase in the weight of the spleen and thymus (200-800 mg/kg) and enhanced the proliferation rates of splenocytes (200-800 mg/kg). TCD and EH-TCD significantly increased the … Show more

“…Similar to results after direct in vitro stimulation of splenocytes, oral administration of protein hydrolysate derived from oyster, tuna cooking drip, salmon, and multiple common carp egg hydrolysates increased ex vivo splenocyte proliferation [ 25 , 39 , 41 , 42 ]. Some of these studies looked into the cell types present in the spleens of these animals in more detail.…”

“…These cells were expected to be both Th1 and Th2 cells, since both Th1 (IL-2, IFNγ) and Th2 (IL-4, IL-5) cytokines were detected in the blood of these animals. These effects might differ between protein hydrolysates, since a tuna cooking drip hydrolysate was found to increase IL-2 and IL-10 [ 42 ], and sensitization with a partial whey hydrolysate increased Treg in the spleen [ 36 ].…”

“…Additionally, in a study analyzing the effect of the enzymatic digestion of tuna cooking drip (CTD) (EH-TCD) on the immune function of Bal b/c mice, the use of TCD and EHTCD significantly increased the production of immunostimulatory cytokines (interleukin 10 and interleukin 2) and increased serum IgG1 and IgG2a levels, and even EH-TCD displayed an immune-enhancing effect stronger than TCD. These results suggest that EH-TCD and TCD might act as immunostimulatory agents to exert immune enhancing effects [ 97 ]. In a later study, after a low molecular weight peptide-NJP (<1 kDa) was obtained from Japanese yellow goby (Nibea japonica) protein hydrolysate, the evaluation of the immunomodulatory effects of this peptide on RAW264.7 cells revealed that NJP was able to promote cell proliferation by affecting cell cycle progression, and it significantly promoted cell phagocytosis, as well as pro-inflammatory cytokine secretion, and promoted IKK α/β by upregulating the protein expression degradation of IκB-α and the activation and translocation of p65 and nuclear factor NF-κB, as well as increasing nitric oxide (NO) synthesis.…”

“…Hou et al [ 15 ] reported that three immunomodulatory peptides (NGMTY, NGLAP, and WT) isolated and purified from Alaska pollock frame hydrolysate had high lymphocyte proliferation activity. Kim et al [ 16 ] reported that tuna cooking drip and its enzymatic hydrolysate not only increased the proliferation rate of mouse splenocytes but also promoted the levels of interleukin (IL)-2, IL-10, and immunoglobulin G (IgG). Yu et al [ 17 ] reported that RVAPEEHPVEGRYLV, a Cyclina sinensis polypeptide, alleviates CTX-induced immunosuppression by enhancing humoral and cellular immunity in mice.…”
