Immunochemical Detection of Tyrosinase in Serum of Patients With Metastatic Malignant Melanomas and From Hardy-Passey Mouse Melanomas

Abstract: Partially purified extracts of soluble Hardy-Passey mouse meldnoma tyrosinase yielded three precipitin lines upon immunoelectrophoresis. Two immunochemically homologous but electrophoretically heterologotis precipitin lines possessed tyrosinase activity whereas a more anodic line was observed which did not display tyrosinase activity.Tyrosinase from serum of patients with metastatic malignant melanomas cross-reacted with antiserum to mouse melanoma tyrosinase, and one precipitin line was seen. “…The neuroblastoma and medulloblastoma lines were supplied by Drs. P. Smith, F. Tomlinson, and .. Kotlowska-Niecko, 1985) and melanoma patients (Del Vecchio and Burnett, 1976) and to inhibit melanisation (Vachtenheim et al, 1986). We recently derived a MoAb (5C12) directed against the carbohydl-ate moiety of ac-M. Lihou (this Institute).…”

“…Later, it was demonstrated to be associated with various human malignancies [6,7]. A serum protein isolated from melanoma patients and studied on immunoelectrophoresis showed some antigenicity in common with mouse tyrosinase [8]. Tyrosinase activity was found in sera of melanoma patients but not in healthy controls [9,10].…”
