Immunodominant Linear B-Cell Epitopes of SARS-CoV-2 Spike, Identified by Sera from K18-hACE2 Mice Infected with the WT or Variant Viruses

Abstract: SARS-CoV-2 surface spike protein mediates the viral entry into the host cell and represents the primary immunological target of COVID-19 vaccines as well as post-exposure immunotherapy. Establishment of the highly immunogenic B-cell epitope profile of SARS-CoV-2 proteins in general, and that of the spike protein in particular, may contribute to the development of sensitive diagnostic tools and identification of vaccine` candidate targets. In the current study, the anti-viral antibody response in transgenic K18… Show more

“…Currently (November 2023), 8,528 linear B-cell epitopes have been submitted to the Immune Epitope Database (IEDB) [44]. The majority of the methods used for linear antibody epitope discovery in SARS-CoV-2 include enzyme-linked immunosorbent assay (ELISA) [36][37][38], peptide arrays [68,69], peptide microarrays [70][71][72][73][74][75][76][77][78][79][80][81][82][83][84], phage display [43,[85][86][87][88][89][90][91][92][93], or bacterial display [94]. The top three SARS-CoV-2 source proteins of linear B cell epitopes in the IEDB data are the ORF1ab (54%), S (30%), and N (9%) proteins [44].…”

“…Our novel vaccine strategy, i.e., taking advantage of both the adjuvant and the tetanus toxoid in a tetanus vaccine, is different from other COVID-19 peptide vaccine strategies published in the literature, i.e., focusing on B-cell and T-cell epitope identification and adjuvant development [2] , [3] , [4] , [5] , [19] , [20] , [21] , [22] , [23] . Furthermore, we used authentic viruses in neutralization assay whereas others did not.…”
