Immunohistochemical Expression of RARα, RARβ, and Cx43 in Breast Tumor Cell Lines After Treatment With Lycopene and Correlation With RT-QPCR

Abstract: Lycopene, the major carotenoid found in tomatoes, is a potent antioxidant associated with the prevention of degenerative diseases such as breast cancer. This effect could be due to the interaction between lycopene and retinoic acid receptors as well as the stimulation of gap junction communication and synthesis of connexin 43. The expression of the RARalpha, RARbeta, and Cx43 proteins was analyzed using immunohistochemistry in two breast cancer cell lines, MCF-7 and MDA-MB-231, and in a fibrocystic dystrophy c… Show more

“…Slides Slides were then counterstained with hematoxylin for 3 min, rinsed in distilled water, and placed on coverslips with an aqueous Faramount mounting media (dako, Glostrup, denmarck). The primary polyclonal antibody was omitted and replaced with PBS as a negative control (39).…”

“…Lycopene appears to have significant benefits in reducing the risk of developing chronic diseases such as cancer and cardiovascular disease [9,10]. The benefits of lycopene could be attributed to not only its antioxidative action but also its capacity to upregulate Cx43 expression in cancer cells [11,12]. The over-expression of Cx43 leads to decreased proliferation in melanoma cells [13].…”

“…Lycopene has recently been shown by immunohistochemical cell staining to specifically increase levels of RARa, and connexin 43 (Cx43) in MCF-7 and MDA-MB-231 breast cancer cell lines, and to strongly increase RARb expression in nontumorigenic MCF10A cells [Chalabi et al, 2007b]. We provide new evidence using methylation specific PCR (MSP) that dietarily relevant, low doses of lycopene (2 lM) can alter DNA methylation of the GSTP1 gene in MDA-MB-468 breast cancer cells, and that this change is coordinated with increased gene expression as suggested by RT-PCR.…”
