Impact of curcumin on sirtuins: A review

Abstract: Curcumin is a bioactive phytochemical that modulates several physiological and cellular processes leading to therapeutic effects against different diseases. Sirtuins are highly conserved nicotine adenine dinucleotide-dependent proteins that regulate the activity of target enzymes and transcription factors by deacetylation. Curcumin possesses both antioxidant and anti-inflammatory properties and has been shown to increase sirtuin-1 (SIRT1) by activating small molecules. Upregulation of SIRT1 by curcumin has bee… Show more

“…In vascular smooth muscle cells, Cur promotes AMPK activation, which enhances superoxide and ATP production. This event stimulates an increase in NAD + levels and SIRT1 activation ( Zendedel et al, 2018 ). Cur-induced SIRT1 upregulation has beneficial effects against a range of diseases including cardiac fibrosis, diabetes, and ischemia/reperfusion injury ( Zendedel et al, 2018 #7; Yang et al, 2013 #5).…”

“…SIRT1 also activates AMPK through LKB1 deacetylation. In fact, AMPK and SIRT1 create a negative feedback loop by affecting each other's activation (37,38). Therefore, a correlation in their expression is expected.…”

“…3). Three of the 4 active compounds (IC 50 <5 μM); curcumin (IC 50 = 2.2 μM), α-mangostin (IC 50 = 1.2 μM), and nobiletin (IC 50 = 0.9 μM), are reported to have SIRT1-activating effects (7,17,28,29,41,43,55). Another compound, HMF showing IC 50 = 1.0 μM, is reported to ameliorate brain and bone injuries (30,31,35,36,45), and recently shown to possess anti-metabolic effects (16).…”
