Impact of sphingosine and acetylsphingosines on the aggregation and toxicity of metal-free and metal-treated amyloid-β

Abstract: The reactivity of sphingosine and acetylsphingosines towards both metal-free and metal-treated amyloid-β is demonstrated showing a correlation of their micellization properties.

“…This difference may indicate that the complex involves two different tautomeric forms of histidine in the peptide's coordination to the Zn 2+ ion. The deshielding effect for His2 may arise from the lower electron density around these protons because of their proximity to the metal 30 . This suggests that the zinc coordination is carried out through Nδ 1 for His2 and through Nε 2 for His3.…”

“…5 The drug development pipeline in AD is undergoing a paradigm shift from single target to multitargetdirected strategies to combat multifaceted amyloid toxicity. [6][7][8] In this context, several groups have designed chemical tools to impede metal-Ab 42 complexation and its associated aggregation-induced toxicity. [9][10][11][12] We have previously developed therapeutic small molecules, peptides and hybrid multifunctional modulators to address multifaceted Ab 42 toxicity.…”

“…2 The accumulation of extracellular senile plaques made up of amyloid-β peptides (Aβ) is the pathological marker of AD. 3,4 Aβ is a normal peptide metabolized in the human brain, which maintains non-β-sheet conformation. 5 Under pathological conditions, such as abnormal cerebral metal ions or misfolded templates, Aβ can transform to β-sheet-rich species, which is the key step to producing toxic Aβ aggregates.…”
