Impairment of the photosynthetic apparatus by oxidative stress induced by photosensitization reaction of protoporphyrin IX

Abstract: Treatment with the herbicide acifluorfen-sodium (AF-Na), an inhibitor of protoporphyrinogen oxidase, caused an accumulation of protoporphyrin IX (Proto IX) , light-induced necrotic spots on the cucumber cotyledon within 12-24 h, and photobleaching after 48-72 h of light exposure. Proto IX-sensitized and singlet oxygen ((1)O(2))-mediated oxidative stress caused by AF-Na treatment impaired photosystem I (PSI), photosystem II (PSII) and whole chain electron transport reactions. As compared to controls, the F(v)/F… Show more

“…Therefore, our results show that the most relevant ROS, which is produced in the chlorophyll-containing lipid membranes of leaf tissue exposed to light, is 1 O 2 both under basal and stress conditions. 1 O 2 oxidizes cellular components, such as nucleic bases (Ravanat et al, 2000), proteins (Davies, 2003), and membranes and this characteristic is exploited in photodynamic therapies to kill cancer cells (Girotti and Kriska, 2004), bacteria (Maisch et al, 2007), and also to conceive herbicides (Fufezan et al, 2002;Tripathy et al, 2007). We have observed that 1 O 2 -mediated LPO precedes cell death in our 1 O 2 -overproducing mutants vte1 npq1 and ch1.…”

“…This consequence may be attributed to the fact that damages in the photosynthetic system reflect on the reduction of the levels of chlorophyll and carotenoids (Santos et al, 2011). As the oxidative stress increases in function of time of exposure to the light, the thylakoids are damaged and lose their capacity to do photosynthesis due to damage in the photosynthetic apparatus (Tripathy et al, 2007), which can explain the smaller liquid photosynthesis observed in plants treated with carfentrazone-ethyl.…”

“…Among responses that plants have against these stresses include increased membrane peroxidation, which affects the structure and function of the photosynthetic apparatus, inactivating the reaction centers of the photosystems (Tripathy et al, 2007).…”
