Improved agarose electrophoretic method for separating alkaline phosphatase isoenzymes in serum.

Abstract: A modified agarose electrophoretic system for the separation of alkaline phosphatase (ALP, EC 3.1.3.1) isoenzymes is described. Bone, liver, high-molecular-mass, and intestinal ALP are separated with high reproducibility. The sensitivity of the agarose system is superior to cellulose acetate in detecting high-Mr ALP. Correlation is good between bone ALP fractions scanned before and after treatment with neuraminidase. Immunoglobulin-bound ALPs, the ALP-lipoprotein-X complex, and the additional ALP fraction obse… Show more

“…1-3 TH is considered to be present when serum ALP is >1000 IU/L; 1 furthermore, the elevation is often accompanied by elevation of both liver and bone ALP isoenzymes. 4,5 We report the case of an infant with Epstein-Barr virus (EBV)-associated TH (EBVATH).A male infant was found to have hepatomegaly at his routine health check-up at 18 months of age, and was referred to hospital by his physician. On physical examination, he was in good general condition, but the liver was palpable 5 cm below the right costal margin, and the spleen was enlarged to 3 cm below the left costal margin.…”

“…Bone alkaline phosphatase activity was quantified by means of agarose electrophoresis with reagents from Analis (Suarl ee, Belgium). (33) Osteocalcin, carboxy-terminal collagen cross-links (CTX), and serum type 1 procollagen (P1NP) were measured on a Modular EE instrument (Roche Diagnostics, Vilvoorde, Belgium) using reagents from the same manufacturer.…”

“…It was recently reported that this band is HIALP. In 1988 Van Hoof et al (1) employed neuraminidase treatment and polyclonal and monoclonal antibodies against the intestinal type and confirmed that the band overlapping bone ALP on agarose electrophoresis is an intestinal variant of ALP. In 1998 Hoshino et al (2) confirmed that this band is a high-molecular band by polyacrylamide gel electrophoresis (PAGE), and Matsushita et al (3) reported that the band could be separated from bone ALP by conversion to ALP5 by subtilisin treatment.…”
