Improved in vitro micropropagation method with adventitious corms and roots for endangered saffron

Abstract: The objective of this study was to investigate development of an efficient in vitro tissue culture system for saffron (Crocus sativus L.) complete with roots and corms. In indirect organogenesis, Murashige and Skoog (MS) media with 3% (w/v) sucrose, 100 mg L−1 ascorbic acid, and the combination of 0.25 mg L−1 2,4-dichlorophenoxyacetic acid (2,4-D) and 1 mg L−1 6-benzylaminopurine (BAP) were best for callus initiation and growth while 1.5 mg L−1 BAP was excellent for high rate of adventitious shoot formation. 1… Show more

“…MS medium containing BA (13.32 µM L -1 ) with 2,4-D (0.90 µM L -1 ) produced 100% callus response in S. chirayita from root explants after five weeks of culture [22]. Similarly, in saffron least concentration of 2,4-D (0.25 mg L -1 ) in combination with and BA (1 mg L -1 ) was found to be best for callus initiation [23]. In several other Gentianaceae species like Swertia angustifolia and Swertia mussotti higher concentration of 2,4-D (3.0 mg L -1 ) and Kn (2.5 mg L -1 ) exhibited best callusing in leaf explants [12,13].…”

“…In addition, the vigorous roots are also generated in ½MS medium supplemented with 0.5 mg dm -3 IBA (Zang et al 2017). However, high induction rate of adventitious roots could be achieved by using MS media supplemented with 2.0 mg dm -3 IBA in an efficient in vitro tissue culture system for Crocus sativus (Zeybek et al 2012). Therefore, different types of medium (MS or ½MS) played distinct roles in plant rooting.…”

“…Besides, cormlet development in Crocus tissue cultures occurs under various PGR treatments. Several papers report the importance of IBA (Zeybek et al, 2012), while others underline the use of (Sharma et al, 2008) for C. sativus. The use of high sucrose concentration and high cytokinin/auxin ratios was proven to be beneficial for plant regeneration and cormlet production from corm-derived somatic embryos in C. vernus (Sivanesan et al, 2012(Sivanesan et al, , 2014.…”
