Improvement of Production of Kojic Acid by a Mutant Strain Aspergillus oryzae, MK107-39.

Abstract: A strain designated MK107-39, producing kojic acid with a high yield, was obtained by a new screening method using a 96-well microtiter plate after NTG treatment of Aspergillus oryze ATCC 22788. The amount of kojic acid produced by strain MK107-39 in a shaking flask was 28 g/l from 100 g/l of glucose, which was 7.7-times higher than that produced by parent strain. The kojic acid yields per cell and the amount of glucose consumed were 9.8 and 6.0-times higher than those of the parent strain. Based on difference… Show more

“…After harvesting, the resultant biomass, including the medium, was centrifuged at 10,000 rpm for 20 min at 4 °C. The biomass was separated and the remaining filtrate was extracted using ethyl acetate (1:1 v/v), and collected aqueous ethyl acetate was concentrated in-vacuo in rota-evaporator at 45 °C to afford 9.0 g as brown oily crude extract (Futamura et al, 2001[8]). The obtained unique brown organic extract was applied for further biological screening.…”

“…Hence ample amount of kojic acid and dry mass were obtained with ethyl acetate extract as the fermentation process progressed up to 13days. Such type of performance of kojic acid biosynthesis and dry mass by A.flavus was reported [50][51][52][53][54][55][56] .…”

“…Although this may not be directly suitable for industrial application, the antibody productivity by A. oryzae can be further improved by several rounds of mutagenesis and additional genetic modi cations such as deletion of other protease genes, optimizations involving protein secretion and the increasing of copy-number for heavy and light chains. Moreover, a further enhanced productivity is also expected with the controlled growth environment employing a bioreactor as previously reported in A. oryzae [41], which will meet the industrially competitive level.…”
