Improving Bioavailability of Hydrophobic Prodrugs through Supramolecular Nanocarriers Based on Recombinant Proteins for Osteosarcoma Treatment

Abstract: Supramolecular nanodrug assembly driven by supramolecular chemistry is becoming a powerful strategy for medication. The potential of engineered proteins as building blocks for nanoformulations is rarely investigated. Here, we developed a new generation of recombinant proteinbased nanodrug carriers, which is very efficient for loading and delivering the hydrophobic prodrug aldoxorubicin. Significantly enhanced anti-tumor effects in osteosarcoma (OS) models were observed. The half-life of the nanodrug reached al… Show more

“…Meanwhile, a series RESEARCH ARTICLE of surfactants containing 3,4-dihydroxylphenylalanine (DOPA) or azobenzene (Azo) moieties were synthesized (Figure 1B The SUP-surfactant coacervates were prepared by electrostatic complexation of SUPs and the corresponding surfactants. [23,24] Typically, SUPs and surfactants were mixed in an aqueous solution with the initial molar ratio of lysine or glutamic acid to surfactant at 1:1. As a result, the solution became turbid and after centrifugation a viscous SUP-surfactant complex was obtained.…”

“…84 In this regard, recombinant proteins or peptides with low immunogenicity have been designed and used for the preparation of photo-responsive hydrogels. 85,86 Recently, Wei and coworkers developed a new type of injectable NIR lightresponsive hydrogel consisting of elastin-like polypeptide (ELP), chitosan, and Au/Ag hybrid nanoparticles (Ag 3 AuS 2 NPs) via electrostatic interactions and forming amide bonds (Fig. 9C).…”
