Improving TRAIL-induced apoptosis in cancers by interfering with histone modifications

Abstract: Epigenetic regulation refers to alterations to the chromatin template that collectively establish differential patterns of gene transcription. Post-translational modifications of the histones play a key role in epigenetic regulation of gene transcription. In this review, we provide an overview of recent studies on the role of histone modifications in carcinogenesis. Since tumour-selective ligands such as tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) are well-considered as promising anti-tumou… Show more

“…Mutations in the TRAIL signaling pathway confers drug resistance and reduces apoptosis in various tumors [30]. In fact, TRAIL driven signaling holds many opportunities and strategies to the development of suitable therapeutics that can trigger apoptosis in TRAIL-resistance tumors [31]. Investigations have revealed possible proapoptotic and anti-apoptotic proteins that regulate the TRAIL pathway and could be implemented as possible drug intervention strategies that can help in targeting anti-apoptotic proteins [32].…”

“…Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) belongs to the tumor necrosis factor (TNF) subfamily, and has attracted substantial attention since it induces apoptosis in tumor cells with nontoxicity towards normal cells (Zhang et al, 2020). As a promising candidate for cancer therapy, TRAIL mainly activates extrinsic apoptotic cell death through interaction with proapoptotic death receptor 4 (DR4) and/or death receptor 5 (DR5).…”
