While many epidemiological models were proposed to understand and handle COVID-19 pandemic, too little has been invested to understand human viral replication and the potential use of novel antivirals to tackle the infection. In this work, using a control theoretical approach, validated mathematical models of SARS-CoV-2 in humans are characterized. A complete analysis of the main dynamic characteristic is developed based on the reproduction number. The equilibrium regions of the system are fully characterized, and the stability of such regions is formally established. Mathematical analysis highlights critical conditions to decrease monotonically SARS-CoV-2 in the host, as such conditions are relevant to tailor future antiviral treatments. Simulation results show the aforementioned system characterization.

Regulatory T cells (Treg) are suppressor cells that control self-reactive and excessive effector conventional helper T cell (Tconv) responses. Breakdown of the balance between Tregs and Tconvs is a hallmark of autoimmune and inflammatory diseases. Due to the positive dependency of both populations on Interleukin-2 (IL-2), it is subtle leverage to restore the healthy immune balance. By employing a mechanistic mathematical model, we studied the IL-2 therapy in order to increase and stabilize Treg population and restrict inflammatory Tconv response. We introduced an adaptive control strategy to design the minimal IL-2 dosage. This adaptive strategy allows for an individualized therapy based on the feedback of immune kinetics of the patient. Our in silico results suggest that a minimal Treg population is required to restrict the transient side-effect of IL-2 injections on the effector Tconv response. The combination of IL-2 and adoptive Treg transfer therapies is able to limit this side effect in our simulations. Implications of our in silico results are discussed in the context of autoimmunity and transplantation.

<p>Actualmente la diabetes mellitus es un problema de salud pública mundial. En este trabajo, se proponen estrategias de control para mantener los niveles de glucosa en sangre de pacientes diabéticos tipo 1 en los rangos ideales en pro de la salud del paciente y su calidad de vida. La primera estrategia propone una retroalimentación de estados con restricciones de positividad, que en términos médicos representa la eliminación de episodios de hipoglucemia durante períodos prolongados de ayuno. Posteriormente, se realiza una extensión para lograr el rechazo de las perturbaciones por ingesta de alimentos, mediante el acoplamiento de un control proporcional, integral y derivativo. La segunda estrategia es un control predictivo con entrada impulsiva y regulación hacia una zona objetivo. Finalmente, el desempeño de las estrategias es evaluado en 50 pacientes virtuales extraídos de la literatura y en el Simulador UVa / Padova aprobado por la Food and Drug Administration de EEUU.</p>
