In vitro and in vivo studies of osteoblast cell response to a titanium-6 aluminium-4 vanadium surface modified by neodymium:yttrium–aluminium–garnet laser and silicon carbide paper

Abstract: The effects of neodymium:yttrium-aluminium-garnet (Nd:YAG) laser and silicon carbide (SiC) paper on the surface micro-topography of titanium-6 aluminium-4 vanadium (Ti6Al4V) alloy were examined in relation to the response of bone cells. The study was performed in three distinct stages: (1) after surface treatment of samples by laser and SiC paper, the surface hardness, surface roughness, corrosion resistance and surface tension were evaluated; (2) the growth of mouse connective tissue fibroblast cells (L-929) … Show more

“…Also, it has been demonstrated that for primary bovine osteoblasts, the wettability is one of the key factors. In our studies , 2007; Khosroshahi et al, 2009), it is shown that the wettability of the surface can provide a better spreading condition for osteoblast cells due to reduced contact angle. Bearing in mind that the adhesion of bone cells to implant surface consists of two stages.…”

“…[20,21] The MTT reagent reacts with the tetrazolium ring (produced by the mitochondrial dehydrogenases of living cells) to produce a blue formazan product, which can be measured spectrophotometrically. The amount of formazan produced is proportional to the number of viable cells present.…”

“…The results from three individual experiments were averaged and the percentage of cell viability was calculated. [20,21] We also evaluated initial and long-term cell attachment for 1 day up to 7 days using MTT assay. Results were calculated as the percentage of cell attachment by its optical density (OD) values.…”
