Kaffir lime has medicinal properties for the treatment of many diseases. We used in vitro culture techniques to maintain quality and increase the production of active compounds in kaffir lime. The objective of this study is to determine the phenotype and genotype of kaffir lime callus cultures grown on media with the addition of 2,4-Dichlorophenoxyacetic (2,4-D) and Benzylaminopurine (BAP) on three generations (G0, G1, and G2). The callus was induced and subcultured on Murashige and Skoog medium added with 2,4-D: BAP at concentrations of 1:0.5; 2:0 and 5:0. Results showed no significant difference in callus initiation time in all treatment groups. Morphological characteristics, including color, texture, and biomass, varied among growth regulator concentrations and generation level. A high level of callus generation corresponds to a more friable texture and a more yellowish callus. Callus grown with the addition of 2,4-D and BAP 2:0 and 5:0 showed a more friable structure and yellowish color than 1:0.5. Growth regulator concentrations in each generation did not affect the callus growth curves, but the length of each phase between generations was different. The exponential phases of G1 and G2 were faster than that of G0. Despite slight differences in phenotype, the DNA profiles of callus suggest the same pattern between treatment groups, thereby indicating that our kaffir lime callus has genetic stability and that the callus can be used as raw material for medicinal purposes. In Thailand, cassava (Manihot esculenta Crantz ‘Pirun 2’) is an important economic crop. The tissue culture technique is used to produce true-to-type cassava plantlets on a large scale within a short period. Therefore, the objective of this study was to investigate the optimum culture medium for cassava axillary bud in vitro propagation. Stem nodes were treated with a solution of 20% (v/v) commercial bleach (25% w/v NaOCl) for 10 minutes. Then, the sterilized buds were cultured on three different types of culture media: Murashige and Skoog (MS), Driver and Kuniyaki Walnut (DKW), and MS supplemented with NAA and BA. Various parameters were evaluated for their effects on plantlet development (height of shoot and numbers of leaves, roots, and shoots after 1 month). The results showed that the mean (± standard deviation) numbers of leaves (6.67 ± 0.82) and roots (2.50 ± 0.55) were significantly higher when they were cultured on MS medium. In addition, the highest number of shoots (3.67±0.82) was obtained on MS medium supplemented with 0.05 mg/l NAA+1 mg/l BA. The cut shoots of plantlets cultured on MS medium without hormones developed into mature plantlets after 1 month. Thus, the MS medium was the optimum for mature cassava Pirun 2 culture. This protocol would be useful for plantlet induction on MS medium supplemented with NAA and BA hormones to produce more shoots and for transfer to MS medium without rooting hormones. Moreover, in vitro derived plantlets were successfully acclimatized on a peat moss which gave the high percentage of transplanting survival (100%). Sipahutar pineapple is very popular in North Sumatra, because of the distinctive sweet taste and normal water content. Furthermore, it is large in size and has a yellow fruit skin color with greenish tips. However, the problem with Sipahutar pineapple production is the limited amount of good quality seeds. The utilization of in vitro culture techniques on pineapples allows the emergence of somaclonal variations, especially in plantlets that have undergone the subculture stage. This somaclonal variation is one of the problems in commercial seedling production, therefore, it is necessary to conduct an initial examination using the inter simple sequence repeat (ISSR) molecular marker. This study aimed to determine the genetic and primary stability of ISSR, which can be used in pineapple plants native to Sipahutar. The methods used include sample preparation, DNA isolation, primer optimization, ISSR primer amplification by PCR method, and electrophoresis. Furthermore, a total of 15 samples were amplified with six ISSR primers, and the data were analyzed by cluster method using the NTSYS-PC software. The final result was visualized in a dendrogram and analysis of diversity was conducted using GenAlex. The results showed that the level of genetic variability of the Sipahutar pineapple, which has undergone in vitro culture using six ISSR molecular markers was 76-97%. Meanwhile, the genetic variability level of the native to Sipahutar pineapple can be influenced by the long culture period and the use of N6-benzyladenine. The primers can be used to observe the genetic variability, except for ISSR 25 with a PIC value of 0.000.
