In-Vitro Carboxymethylation of Low Density Lipoprotein Alters its Metabolism Via the High-Affinity Receptor

Abstract: Carboxymethylation of lysine residues has been shown to result from oxidation of glycated proteins in vivo and in vitro leading to an augmentation of proteins' net negative charge. The metabolism of carboxymethylated low density lipoprotein (LDL) was studied in cultured human fibroblasts and mouse peritoneal macrophages. In vitro carboxymethylation was achieved by incubation of LDL with glyoxylic acid in the presence of Na(CN)BH3. Carboxymethylation inhibited metabolism of LDL via the high affinity receptor in… Show more

“…33,34 It is relevant that methylation of LDL abolished its recognition by LDL receptors, 35 retarded the degradation of aggregated LDL by macrophages, 36 and decreased CE formation in macrophages. 37 It is possible that hyperhomocysteinemia may inhibit lipid or protein methylation in LDL, which may result in increased endocytosis of LDL-derived CE in the lesions.…”

“…Further studies are needed to clarify the epitope structure of this antibody. Because CML does not undergo receptor recognition (56), it seems likely that non-CML structure(s) generated in GA-BSA may serve as a signal for the receptor recognition. Aldoamine, an Amadorirearranged GA adduct, has been shown to form Schiff base linkages with another amino group, leading to covalent cross-linking of proteins (17,18).…”
