In vitro culture of human osteosarcoma cell lines: a comparison of functional characteristics for cell lines cultured in medium without and with fetal calf serum

Abstract: Serum-free culture media can be used for in vitro studies of several osteosarcoma cell lines, but the optimal medium varies between cell lines and thus depends on: (i) the cell lines to be investigated/compared; (ii) the functional characteristic that is evaluated (proliferation, cytokine release); and (iii) whether coculture experiments are included.

“…26 Interestingly, CCL2 has been described previously as the predominant chemokine secreted by Cal72 cells. 27 Our in vitro data show that the Slo knockdowninduced decrease of CCL2 mRNA correlates with a reduction in CCL2 secretion. In our OS xenograft model, this reduction in CCL2 secretion was accompanied by a marked decrease in macrophage infiltration, while the level of NK cells was unchanged.…”

“…In some cancers, the presence of natural killer cells, eosinophils, granulocytes and B cells has also been reported. 34 OS cells are known to produce numerous chemokines 27,35 and to express a high level of BK channels 12 which are, interestingly, implicated in different steps of the secretary process. 9,10,36,37 These observations led us to explore a putative involvement of BK channels in chemokine production.…”

“…However, the animal serum contains undesired source of xenogenic antigens and bears the risk of transmitting animal viral, prion and zoonose contaminations. 63,64 Hence, the proposed criteria for cGMP require that animal-derived supplements should, whenever possible, be removed or replaced with synthetic recombinant supplements. In contrast to animal serum, the use of autologous serum is advantageous because it eliminates the risk of infectious disease transmission completely.…”

“…This medium, which was developed for the in vitro production of TILs under safety conditions, has only been described previously as providing a serum-free environment for various previously established tumor cell lines [distributor's information, Sezary syndrome, osteosarcoma (Bruserud et al 2005)]. The cell lines directly established and grown in X15 medium are particularly interesting for clinical use; since these cells have never been in contact with FCS, there is no risk that they have been contaminated by pathogens, such as viruses and prions, originating from FCS.…”
