In Vitro Evaluation of Gd3+-Anionic Linear Globular Dendrimer-Monoclonal Antibody: Potential Magnetic Resonance Imaging Contrast Agents for Prostate Cancer Cell Imaging

Abstract: These results showed that Gd(3+)-ALGDG2-C595 is a potential prostate molecular imaging agent and could be considered as an ideal functional nanoprobe. Additionally, further investigations by clinical trials are in the pipeline.

“…ALGDG2 was introduced for the first time by Namazi and Adeli (2005) 19 . Easy synthesis, low molecular weight, well-known functional groups on the surface of the spherical particle, monodispersity, high purity and hydrophilicity, increased permeability to cancerous cells and most importantly low toxicity with low immune system stimulation when injected locally 20 for its biocompatible PEGylated core and biodegradable sidelong citric acid groups made ALGDG2 a suitable carrier for a range of drugs and agents, such as anticancer 21 , antiviral 22 , antibacterial drugs 23 and imaging agents 24 .…”

“…This dendrimer consists of a biocompatible PEGylated core and biodegradable peripheral citric acid groups which its monomeric units are the chemical intermediates or products of metabolic pathways (such as, Krebs cycle) [27]. These compounds can be metabolised and removed by the cellular processes or used as cells' energy source step by step.…”

“…Another contribution to the topic has been conducted by Mohammadzadeh et al [54] as they developed a selective nano-theranostic system composed of AS1411-functionalized anionic linear globular dendrimer G2 (ALGDG2) for the specific delivery of Iohexol to breast cancer cells. ALGDG2 is generally considered as an ideal carrier for a broad spectrum of anticancer, antiviral, and imaging agents due to its low molecular weight, low toxicity, immunocompatibility, biodegradability (because of its sidelong citric acid groups), high purity and hydrophilicity, monodispersity, favorable permeability to cancer cells, and possession of various wellknown functional groups on the spherical particles' surface [55][56][57][58]. Taken together, these nano constructs exhibited a considerable potential for reducing the number of cancer cells by maximizing the accumulation of iohexol in the tumors while they minimized the toxicity of Iohexol on normal cells [54].…”
