In vitro Fertilization with Isolated, Single Gametes Results in Zygotic Embryogenesis and Fertile Maize Plants

Abstract: We demonstrate here the possibility of regenerating phenotypically normal, fertile maize plants via in vitro fertilization of isolated, single sperm and egg cells mediated by electrofusion. The technique leads to the highly efficient formation of polar zygotes, globular structures, proembryos, and transition-phase embryos and to the formation of plants from individually cultured fusion products. Regeneration of plants occurs via embryogenesis and occasionally by polyembryony and organogenesis. Flowering plants… Show more

“…Egg cells and sperm cells were isolated from ears and pollen of this line as described by Kranz and LoÈ rz (1993). Electrofusion (in vitro fertilisation) of single gametes and cultivation of in vitro zygotes was performed according to methods described in the same paper.…”

“…The cells were collected and stored at A80°C until usage. After gamete fusion, in vitro zygotes were cultivated for 18 h without feeder cells in the medium described by Kranz and LoÈ rz (1993) to avoid RNA contamination from lysed feeder cells.…”

“…Novel breeding strategies that will allow improvement of genetic stocks need to be developed. In vitro fertilization (IVF) leads to the production of hybrids between plants that do not normally cross (Kranz and Lörz 1993;Faure et al 1994). Pollen transformation followed by IVF may enhance the efficiency of genetic transformation by taking advantage of the sexual reproduction route, resulting in development of stable, novel genotypes of economically superior trees.…”

“…Since the first successful in vitro fertilization in angiosperms (Kranz et al 1991;Kranz and Lörz 1993) great advances have been made in this field (see Theunis et al 1991;Kranz and Dresselhaus 1996;Yang and Zhou 1998;Faure 1999). Most of the work has, however, been done on monocots, e.g.…”
