In Vitro Propagation of Agave americana by Indirect Organogenesis

Abstract: Factors such as slow growth, low rates of sexual and asexual reproduction, and viability of seeds among others limit the massive propagation of Agave americana L. by conventional methods. In this study, callus induction and shoot proliferation was determined in A. americana using Murashige and Skoog (MS) medium supplemented with dicholorophenoxyacetic acid (2,4-D) and 6-benzyl adenine (BA). Meristematic tissue was used as the explants, and were plac… Show more

“…The highest number of shoots of Aloe vera was obtained in medium containing BAP comparison with another cytokinins (Lavakumaran and Seran, 2014). Agave Americana planted on MS medium supplemented with BAP had the highest number of shoots per callus after 4 weeks from culturing (Lecona-Guzmán et al, 2017). Shoots regenerated of Agave angustifolia from shoot tip on the media with 0.4 mg/l produced the best number of shoots.…”

“…For callus growth in a liquid medium, two strategies were used. The first strategy consisted of growing the selected callus in the same callus induction treatment, and the second strategy consisted of using combinations of hormones: the first treatment was using the combination of 2,4-D + 6-Benzylaminopurine (BAP) (0.5 + 9 mg L −1 ) [16], and the second treatment was using the combination of naphthaleneacetic acid (NAA) + BAP (0.5 + 0.5 mg L −1 ) [17]. Cell viability was determined using the Trypan blue technique [18].…”
