In vitro selection and identification of sweetpotato (Ipomoea batatas (L.) Lam.) plants tolerant to NaCl

Abstract: In vitro selection of sweetpotato (Ipomoea batatas (L.) Lam.) plants tolerant to NaCl was achieved using embryogenic suspension cultures of sweetpotato cv. Lizixiang and gamma-ray induced mutation. Cell aggregates from embryogenic suspension cultures of Lizixiang were irradiated with 80 Gy gamma-ray, and 1 week after irradiation they were cultured in a selective medium containing 342 mM NaCl for in vitro selection. A total of 276 plants were regenerated from the irradiated 2,783 cell aggregates by a two-step i… Show more

“…The results showed also that carotenoids were highly increased under salinity stress. This may be due to adaptation of the plant with high salt stress through stimulation of antioxidant defense systems He et al, 2009). Carotenoids are considered as one of the most important non-enzymatic antioxidant which induced decrease the accumulation of ROS under salt stress (Mittler et al, 2004) also, they induce abscisic acid hormone that promotes stomatal closure when exposed to stress (Leung & Giraudat, 1998).…”

“…A useful in vitro protocol for salt resistance using different concentrations of NaCl as selection tools has also been applied to many crops including sugarcane, potato, sweet potato, and beet (Prakash and Jack, 1993;Yang et al, 2005;Gandonou et al, 2006;He et al, 2009).…”

“…In vitro culture is also an effective way to select fairly drought- (Sorkheh et al 2010) and salt- (He et al 2009) tolerant genotypes that could be grown in harsh environmental conditions, as is the case for pistachio growing in Algeria (Benmahioul et al 2009a). Despite advances in pistachio micropropagation (Barghchi and Alderson 1989), the results are still not as good as those achieved with other fruit species.…”
