IncX3 Epidemic Plasmid Carrying bla NDM-5 in Escherichia coli from Swine in Multiple Geographic Areas in China

Abstract: Six imported pigs originating from Guangdong, Henan, and Hunan provinces in China during October 2015 to February 2017 were cultured and found to be positive for meropenem-resistant The samples yielded 9 isolates of diverse sequence types carrying on IncX3 (8 isolates from 5 farms) or IncFII (1 isolate from 1 farm) plasmids. The gene was coharbored by 4 isolates. The IncX3 plasmids (∼46 kb) carrying were identical or nearly identical to each other.

“…The IncX3-type plasmid appears to be an efficient vector for carbapenemase genes, as it has been reported to carry the bla NDM , bla OXA-48 -like, and bla KPC genes (25)(26)(27) and can disseminate the genes both inside (28) and outside (29,30) of the clinical setting. IncX3 plasmids also harbor the hns gene, a homolog of which was reported to allow plasmids to be transferred to a bacterial host by minimizing their fitness cost (31).…”

“…[7][8][9] These plasmids are known to disseminate various carbapenemase genes, including bla KPC and bla NDM . 10,11 To date, only two OXA-181-producing E. coli isolates have been reported in China, namely, in Sichuan and Henan. 12,13 Here, we present the first report of the identification of a K. pneumoniae isolate harboring bla OXA-181 in China.…”

“…In 2012, our group reported the involvement of IncX3 plasmids (represented by pNDM-HN380) in the dissemination of bla NDM-1 in multiple geographic areas in China ( Ho et al, 2012b ). Subsequently, pNDM-HN380-like plasmids carrying bla NDM-1 or variants have also been described in India, the Arabian Peninsula, Europe, and Australia ( Wailan et al, 2015 ; Petrosillo et al, 2016 ; Riazzo et al, 2017 ; Choudhury et al, 2018 ; Ho et al, 2018a ). In a recent survey of more than a thousand carbapenem-resistant Enterobacteriaceae originating from hospitals located in 25 provinces and municipalities in China, the prevalence of bla NDM was found to be 31% ( Zhang et al, 2017 ).…”
