Depression is declared the second leading cause of disability worldwide. Recently, cases of depression have increased significantly in adolescents, young adults as well as in elder population. Monoamine oxidase‐A (MAO‐A) is considered one of the major targets for the treatment of depression. In the current study, we have designed and synthesized various indole functionalized piperazinyl derivatives and evaluated them for in vitro MAO‐A inhibitory activity and in vivo antidepressant‐like activity. Most of the compounds were found to possess potent MAO‐A inhibitory activity with IC50 values in the sub‐micromolar range along with significant selectivity over MAO‐B. Compounds RP1 and RP9 emerged as the most promising reversible MAO‐A inhibitors with IC50 values of 0.11±0.03 μM and 0.14±0.02 μM and displayed selectivity of 193 folds and 178 folds over Monoamine oxidase‐B (MAO‐B), respectively. In the series, RP1 showed good intracellular ROS inhibitory activity along with neuroprotective properties. These compounds were found nontoxic against SH‐SY5Y cells and explored antidepressant activities. In the in vivo Forced swimming test (FST) and Tail suspension test (TST) studies, RP1 exhibited potential antidepressant‐like behavior similar to standard drug fluoxetine while compound RP9 showed antidepressant‐like activity only in the TST studies. The molecular docking and dynamics studies further supported the results obtained in the in vitro and in vivo studies. Thus, the indole functionalized piperazinyl derivatives were found to be promising ligands and can be developed as new antidepressant molecules.

Alzheimer’s disease is a neurological disorder that progressively affects the brain cells. It is sometimes accompanied with dementia and commonly seen in elderly people. It ultimately leads to impaired cognitive functions and loss of memory. Widely used drugs such as donepezil and galantamine successfully abate mild to moderate symptoms of the disease though they have profound side effects. Natural products have been a source of medicine for over thousands of years. Plant secondary metabolites such as alkaloids, flavonoids, terpenoids, etc. are major sources of such medicines. In recent times, the use of natural products to treat various diseases has gained a lot of popularity. Extensive research is being undertaken to identify and isolate various natural products that can be used to not only reduce the disease symptoms but also as a permanent cure. The review article aims to summarize the effects of various natural products and their neuroprotective mechanisms that can be helpful to cure Alzheimer’s disease.

Mental disorders are commonly occurring and often seriously impairing in many countries throughout the world. The total number of people suffering from anxiety or depression amount to 615 million, Close to 10% of the world's population is affected. Chrysanthemum indicum is an ancient herbal medicine used as an antiarthritic, anti-inflammatory and anti-anxiety in many Asian countries. The elevated plus-maze (EPM) and open field test has been most familiar tool in the investigation of the neurochemical transmission basis of anxiety, for testing anxiolytic drugs. The anxiolytic activities of Ethanolic and Chloroform extracts of Chrysanthemum indicum Linn flowers were evaluated using the elevated plus-maze (EPM) and Open field. The ethanolic extract results are suggestive of anxiolytic activity in dosages around 250 and 500 mg/kg, as expressed by elevation of the time spent on the open arms in the plus-maze; a decrease of freezing and increase of deambulation in the open field test.
