Indole-3-carbinol suppresses NF-κB activity and stimulates the p53 pathway in pre-B acute lymphoblastic leukemia cells

Abstract: B cell precursor acute lymphoblastic leukemia (BCP-ALL) is the most common type of cancer in children. Dramatic improvements in primary therapy for childhood ALL have led to an overall cure rate of 80 %, providing opportunities for innovative combined-modality strategies that would increase cure rates while reducing the toxic side effects of current intensive regimens. In this study, we report that indole-3-carbinol (I3C), a natural phytochemical found in cruciferous vegetables, had anti-leukemic properties in… Show more

“…Both murine disease models are characterized by the induction of harmful inflammatory states that were alleviated through the AHR-dependent promotion of antiinflammatory Treg differentiation. The dietary metabolite I3C has been shown to limit proinflammatory responses through additional mechanisms such as suppression of nuclear factor kB signaling pathways (Safa et al, 2015). Additionally, I3C or its metabolites exhibit antimicrobial properties, such as inhibition of Escherichia coli and Staphylococcus aureus biofilm formation (Monte et al, 2014).…”

“…EGCG induced nuclear accumulation and transcriptional activity of NRF2, as well as binding of NRF2 to the antioxidant response element sequence located at the target gene promoters in human MCF10A breast epithelial cells [142-146]. Indole-3-carbinol purified from the brassica genus of he cruciferous vegetable family ( Brassicaceae ), including broccoli, cabbage, cauliflower, mustard and radish, as reviewed in [147-150]. In acidic environment of stomach, indole-3-carbinol is transformed into diindolylmethane condensation compounds.…”

“…In acidic environment of stomach, indole-3-carbinol is transformed into diindolylmethane condensation compounds. Both indole-3-carbinol and diindolylmethane induced apoptosis in various cancer cell lines by modulating nuclear receptor-mediated signaling [147-150]. Indole-3-carbinol suppresses NF-κB activity and stimulates the TP53-dependent pathway and its downstream target genes encoding TP53 Upregulated Modulator of Apoptosis (PUMA), Phorbol-12-Myristate-13-Acetate-Induced Protein (PMAIP)-1, and Apoptotic Protease Activating Factor (APAF)-1 in pre-B acute lymphoblastic leukemia cells [150].…”

“…In human tumorigenesis, NFKB is an important factor in cancer cell survival [11]. In fact, alterations to the NFKB pathway are particularly recognized in ALL and other leukemias, for example, its regulation should facilitate the apoptosis of leukemic cells [12].…”

“…The molecular signaling pathways associated with the hematopoiesis process are varied but deregulation in one or more points should be decisive in ALL development. Moreover, the cytogenetic pattern is an important determinant of ALL development; for example, t (1,19), t (4,11), t (12,21) and t (9,22) affect the expression of the enzymes that regulate the intracellular signaling process [1]. As we know, immunophenotype, age, race, white blood cell (WBC) count, and gender are important prognostic factors.…”
