“…Mutations affecting the overall lysine content of cereal endosperm have received considerable attention because lysine is the limiting essential amino acid in cereal grains when used as food for monogastric animáis (Mertz et al 1964;Nelson et al 1965;Munck et al 1970;Singh and Axtell 1973;Ingversen et al 1973;Dolí 1973Dolí , 1983. However, these so-called high-lysine mutations do not specifically affect lysine biosynthesis and their effect on lysine content is the indirect result of profound alterations in the pattern of protein accumulation throughout development and, at least in some cases, they also affect the metabolism of carbohydrates and lipids, as well as seed morphology.…”

“…While a major portion of the B hordein structural genes at the Hor-2 locus has been deleted in Ris0 56 (Kreis et al 1983), in the other two mutations, both of which map in sepárate loci of chromosome 7 (synonym 5H; Karlsson 1976Karlsson , 1977, trans-acting regulatory genes seem to be affected . The lys 3a mutation drastically reduces the net accumulation of all hordein polypeptides except D-hordein (Ingversen et al 1973;Hopp et al 1983;Thompson and Bartels 1983;Kreis et al 1984). The reduction in storage protein accumulation seems to bring about an increase in the synthesis of other polypeptides and in the level of free amino acids (Brandt 1976;Shewry et al 1980).…”

“…The high-lysine genes usually decrease the synthesis of the lysine deficient storage protein, hordein (4,11,29,33). Hordein polypeptides are synthesized during endosperm development, deposited in protein bodies and may account for half of the total endosperm protein at seed maturity (7, Abbreviations: CM = Carlsberg Monoclonal; FITC = fluorescein isothiocyanate; lg = immunoglobulin; kD = kilodalton; MAb = monoclonal antibody; p = protein; PAGE = polyacrylamide gel electrophoresis; SDS = Sodium dodecyi sulphate; Tris = tris-(hydroxymethyl)-aminomethane.…”

“…A final viscosity analysis of seeds of M 4 plants led to the selection of four lines for detailed investigations. The M 5 plants from these four lines and the high lysine mutant Riso 1508 (18) were obtained from the Carlsberg Research Farm, Hyidagerg~rd, Denmark.…”

“…Since M-1460 also has enlarged embryos and shriveled endosperms, it is possible that M-1460 might be a high lysine mutant of the Riso-1508 type (18). This possibility was examined by comparing the amino acid composition of total seed protein from Minerva, M-737, M-1460 and Riso 1508 (Table II).…”
