Induction and Regulation of Xenobiotic-Metabolizing Cytochrome P450s in the Human A549 Lung Adenocarcinoma Cell Line

Abstract: Several cytochrome P450 (CYP) enzymes are expressed in the human lung, where they participate in metabolic inactivation and activation of numerous exogenous and endogenous compounds. In this study, the expression pattern of all known xenobiotic-metabolizing CYP genes was characterized in the human alveolar type II cell-derived A549 adenocarcinoma cell line using qualitative reverse transcriptase/polymerase chain reaction (RT-PCR). In addition, the mechanisms of induction by chemicals of members in the CYP1 and… Show more

“…This view was supported by the inability of 2,4,3′,5′-tetramethylstilbene, a P4501A1/1B1 inhibitor, to block anti-B [a]PDE adduct formation (unpublished data Gelhaus and Blair). Other P450's that are expressed in human lung cells include, 2A6, 2A13, 2B6, 2E1, 2C18, 2F1, 2J2, 2S1, 3A5 and 4B1 (56)(57)(58)(59)(60).…”

“…In addition, there may be limited application for this strategy to succeed in treating lung cancer due to difficulties in administering and targeting siRNA. A549 is found to express telomerase and several CYP forms including CYP1A1 and possesses metabolic activities toward xenobiotics [21][22]. Furthermore, A549 cell line is known to retain several characteristics of human lung epithelial cell CYP expression, which makes it a valuable model for mechanistic studies on the induction of the pulmonary CYP system [21][22][23].…”

“…1,15 The lung is a primary site of exposure for inhaled toxins along with carcinogens that can ultimately yield lung carcinomas. 16 CYP1A1 is strongly induced by cigarette smoking. 15 Many compounds, including some found in cigarette smoke, are not hazardous until metabolized by CYP1A1.…”
