Induction of Ovarian Maturation and Spawning in Penaeus monodon Broodstock by Double-Stranded RNA

Abstract: Ovarian maturation in crustacean is under the control of gonad-inhibiting hormone (GIH); a neuropeptide secreted from X-organ sinus gland complex in eyestalks. Unilateral eyestalk ablation that partially destroys GIH source is therefore a general practice in Penaeus monodon hatchery to induce ovarian maturation and spawning. Our previous report showed that silencing of GIH expression by GIH-specific double-stranded RNA (GIH-dsRNA) resulted in an increased expression level of vitellogenin in P. monodon, thus su… Show more

“…monodon to investigate VIH (named gonad-inhibiting hormone: GIH in these reports) function, and as a means of inducing ovarian maturation and spawning on-site in Thailand, where this species is an important target of culture. The results of Treerattrakool et al (2010) show that the use of GIH-dsRNA to create GIHknockdown shrimp, is as effective as unilateral eyestalk ablation in promoting ovarian maturation and spawning. The results are very promising, and appear to be almost at the level of being able to be commercially utilized in the field.…”

“…Recently, Treerattrakool et al (2008Treerattrakool et al ( , 2010 have conducted studies using double-stranded RNA in Pe. monodon to investigate VIH (named gonad-inhibiting hormone: GIH in these reports) function, and as a means of inducing ovarian maturation and spawning on-site in Thailand, where this species is an important target of culture.…”

“…This may pave the way to develop an alternative and powerful technique in place of eyestalk ablation with similar impact. Efforts have been made to silence/knockout this gene using the technique of RNA interference by some workers but the success achieved is not as per the expectations and further research is needed in this area [25,26]. Treerattrakool et al (2011) while working on P. monodon used the technique of RNA interference for inducement of maturation both in wild and captive shrimp and reported that shrimps injected with anti-GIH double stranded (ds) RNA showed advanced maturation [25].…”

“…In recent studies, silencing of GIH expression in Penaeus monodon using GIH-specific double stranded RNA (GIH-dsRNA) increased the expression level of vitellogenin 21,22 . In a subsequent study, single injection of GIH-dsRNA into previtellogenic female P. monodon led to ovarian maturation and spawning in some prawn indicating that this method was still less effective than unilateral eyestalk ablation 23 . Dynamic levels of vitellogenin in the haemolymph were observed during reproductive activities.…”
