Influence of Cultivar and Harvest Time on the Amounts of Isoalliin and Methiin in Leek (Allium ampeloprasum var. porrum)

Abstract: Extracts of 31 leek cultivars were analyzed using liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) to determine the distribution of the two most abundant S-alk(en)yl-l-cysteine sulfoxides (ACSOs) in leek, that is, isoalliin and methiin. The isoalliin concentration of the white shaft and green leaves of the 31 leek cultivars varied from 15 to 53 mg/g dry weight (dw) and from 9 to 45 mg/g dw, respectively, whereas the methiin concentration varied from 3 to 16 mg/g dw and from 1 to 10 mg/g dw, respecti… Show more

“…The extraction of S ‐methylcysteine sulphoxide was performed as described by Bernaert et al. , with the supernatants diluted 1:10 using HPLC grade water containing 0.1% formic acid. Analysis was performed by LC‐MS/MS using an Agilent SB‐AQ 1.8 μm (100 × 2.1 mm) C18 column, with an Agilent Zorbax guard column, connected to a model 1290 infinity 6490 Triple Quad LC‐MS/MS system (Agilent Technologies) with a 6490 mass spectrometer.…”

“…Sturon, although other authors reported a prevalence of isoalliin (Yamazaki et al 2010;Bernaert et al 2012). Starkenmann et al (2011) found a much higher level of isoalliin than alliin in onion tissues.…”

“…Bystricka et al (2015) observed differences between cultivars and a statistically significant increase of quercetin formation in all cultivars during vegetation. Also Bernaert et al (2012) reported differences in S-alk(en)yl-l-cysteine sulfoxide (ASCO) content in various garlic cultivars. Lee et al (2015) showed differences in secondary metabolite content Six onion cultivars Bingo, Dormo, Elenka, Elbrus, Spirit, and Sturon grown in the Czech Republic for commercial purposes were analysed to investigate the content of health-promoting compounds.…”

“…Leek cultivar and tissue had an effect on the S-alk(en)yl-L -cysteine sulfoxides, i.e. isoalliin and methiin amounts (Bernaert et al 2012b ). Cultivars Artico and Apollo F1 rated highest for the mean isoalliin and methiin concentration, respectively.…”
