Moxifloxacin is a broad spectrum fluoroquinolone antibacterial agent. We examined the hepatic redox status and plasma biomarkers of nephrotoxicity and hepatotoxicity in rat following administration of moxifloxacin (MXF). Twenty-four Wistar rats, 180–200 g, were randomized into four groups (I–IV). Animals in group I (control) received 1 mL of distilled water, while animals in groups II, III, and IV received 1 mL each of MXF equivalent to 4 mg/kg b.w., 8 mg/kg b.w., and 16 mg/kg b.w., respectively. After seven days, plasma urea, bilirubin, and creatinine were significantly (P < 0.05) elevated in the MXF-treated animals. Activities of alkaline phosphatase, aspartate aminotransferase, and alanine aminotransferase were significantly increased in the plasma of MXF-treated animals compared to control. Also plasma total cholesterol, HDL-cholesterol, LDL-cholesterol, and triglycerides increased significantly in the MXF-treated groups relative to control. Moreover, MXF triggered a significant decrease in hepatic catalase, superoxide dismutase, and glutathione-S transferase activities. Likewise, MXF caused a decrease in the hepatic levels of glutathione and vitamin C. A significant increase in hepatic MDA content was also observed in the MXF-treated animals relative to control. Overall, our data suggest that the half-therapeutic, therapeutic, and twice the therapeutic dose of MXF induced nephrotoxicity, hepatotoxicity, and altered hepatic redox balance in rats.

Levofloxacin is a third generation of fluoroquinolones family. It is commonly used in the treatment of a several bacterial infections. However, misusing antibiotics impose damage to hepatocytes and myocytes. The present study was conducted to access the effect of levofloxacin on tissue histology in mice taking in consideration dose and duration of exposure. 24 matured male Albino mice were divided into 3 groups, Group G1: was considered as a control group. They received normal saline intraperitoneally / day. Group G2: They received 10.7 mg/kg/day of levofloxacin intraperitoneally for 10 days. Group G3: They received 10.7 mg/kg/day of levofloxacin intraperitoneally for 3 weeks. Microscopic examination of liver sections of group G2 revealed severe congestion of blood vessels in the portal area and central veins with inflammatory cells infiltration. While in group G3, Apoptosis, Degeneration and necrosis of hepatocytes with giant cell transformation were noticed. In addition to kupffer's cell activation. Heart sections showed moderate congestion of blood vessels with edema in between the myocytes and inflammatory cells infiltration. Group G3 Necrosis with pyknosis of cardiac muscle nuclei was noticed. We concluded that levofloxacin induces toxic effects on liver and heart according to the dose of administration and duration of treatment.

Fluoroquinolones are more likely than any other antibacterial drug classes to cause serious side effects. The purpose of this study is to assess the protective effects of vitamin E, curcumin, and ginger in male rats against the side effects of levofloxacin (LFX).Ninety male Wistar rats were placed into six groups, each of 15 animals. Groups 1 and 2 received distilled water or olive oil and kept as normal controls, while groups 3-6 received LFX [10 mg/kg body weight (BW)] alone or in combination with vitamin E (100 mg/kg BW.), curcumin (200 mg/kg BW.) or ginger (200 mg/kg BW.), respectively. All medications were administered orally via gavage once a day for 5 successive days. Samples of blood were collected at zero day , 7 th and 14 th day post-treatments for biochemical analysis, while liver and kidney tissues were dissected and subjected to antioxidants estimation. Levofloxacin administration caused hepatic and renal damage evidenced by significant increase in serum liver enzymes, urea (76.43±3.70, 72.67±0.47, 75.00±6.42), creatinine (2.48±0.01, 2.48±0.01, 2.50±0.01), total cholesterol (166. 67±6.81, 174.00±4.04, 176.00±2.88), triglycerides (269.67±5.84, 289.33± 4.91, 249.67±1.86), and low density lipoprotein-cholesterol (LDL-C) (243.00±7.09, 213.67± 5.03, 226.33±5.45) levels. Meanwhile, significant decline in serum levels of high density lipoprotein-cholesterol (HDL-C) (20. 00±1.11, 23.33±3.38, 20.10±1.53), total protein (5.77±0.03, 5.97±0.09, 5.87±0.09), albumin (3.45±0.03, 3.60±0.06, 3.57±0.09) and A/G ratio (1.49±0.02, 1.51±0.05, 1.55±0.09) were determined in relation to normal control at zero, 7 th and 14 th day post-treatments, respectively in a time dependent manner.
