Influence of estradiol on mammary tumor collagen solubility in DMBA‐induced rat mammary tumors

Abstract: Estradiol plays a vital role in the growth and development of mammary glands. It is a potent stimulator of metabolic processes in normal and carcinoma breast. A critical factor in determining mammary glandular morphology is the stroma. Collagen is a predominant component of the extracellular matrix and cell-collagen interactions are essential carcinogenesis. The present investigation explored the influence of estradiol on collagen solubility and metabolism in mammary tumors during tumor progression and regress… Show more

“…The administration of 17-␤-estradiol increases the serum concentration of the hormone [13]. In the current study, the AIN-93G + E 2 group exhibited relatively low serum concentration of the hormone compared to the other rat groups.…”

“…There is no reference data for the normal concentration of serum E 2 . Additionally, the concentration depends on the hormonal status of the animal (stage of estrous cycle) [12,13], and other external factors to which the animal is exposed such as diet [14,15] and drug treatment(s) [12]. Similar studies -in which E 2 concentration was measuredshowed that the concentration of the hormone in the serum of 15-wk old female SD rats fed AIN-93 G meals was 30.4 pg/ml [16].…”

“…In the present study 7,12-dimethylbenz[a]anthracene (DMBA) a chemical carcinogen was used for the development of mammary tumours in rats (Female SD /Wistar) rats [13,14,16]. This model mimic's breast cancer pathology closely as the active metabolites have the capacity for damaging DNA molecules which is a main initiating event in mammary cancer progression [17][18][19][20]. DMBA is highly carcinogenic and lipophilic but requires metabolic activation for its carcinogenicity.…”

“…Along with oxidative stress, recent investigations also depicted 56 the adverse effect of ER stress on male sex organ and fertility in diabetic 57 condition [15]. ER is a eukaryotic cellular organelle present within the 58 cytoplasm and formed by the interconnected network of a series of 59 folded membranes (continue its expansion with the outer membrane 60 of the nucleus) and flattened sacs. SR (sarcoplasmic reticulum) is a 61 type of ER which acts mainly as a store of Ca 2+ and helps pumping it 62 out on stimulus response.…”

“…Along with these alterations, reduced 515 sperm count, motility and increased sperm abnormality were also 516 observed in the diabetic group. Decreased testis-to-body weight ratio 517 is associated with the degradation of the structural proteins [59]. SDH 518 catalyses conversion of sorbitol to fructose and provide energy to the 519 sperm cells to carry out normal metabolism, whereas G6PD is involved 520 in the glutathione metabolism and helps in the hydroxylation of 521 steroids by providing reducing equivalents [60].…”
