Steady state levels of hypothalamic expression of the genes encoding corticotropin-releasing hormone (CRH), proopiomelanocortin (POMC), arginine vasopressin (AVP), and oxytocin (OT) were studied in rats to investigate the mechanisms underlying the transitions between hypercorticalism during lactation and normocorticalism upon weaning. During lactation, CRH mRNA levels and blood titers of adrenocorticotropin (ACTH) were found to be significantly reduced, although POMC mRNA levels in the anterior pituitary were not significantly different from those found in cycling virgin (control) rats; during all phases of lactation, an inverse relationship was observed between the blood levels of ACTH and corticosterone (CORT). Plasma prolactin (PRL) concentrations were elevated 30-fold during lactation. Whereas steady state levels of OT mRNA were markedly increased throughout lactation, those of AVP mRNA were only transiently (initially) elevated, and the blood levels of these hormones were not significantly altered in lactating as compared with cycling virgin and postlactating rats. CRH and POMC gene expression and blood levels of ACTH, CORT, and PRL were normalized within 1-3 d of removal of suckling pups. The temporal relationships between the biosynthetic profiles of the various peptide hormones and the patterns of ACTH and CORT secretion during the two physiological states suggest that lactation-associated hypercorticalism does not merely result from increased ACTH secretion; although still not well substantiated at this time, the evidence points to contributory roles of PRL, OT, and AVP in the hypercorticalismic state found during lactation. The aim of these experiments was to investigate the effects of psychological stress and oxytocin treatment on levels of norepinephrine, dopamine and serotonin in the hypothalamus, hippocampus, striatum, midbrain and brainstem of lactating females. Stress and oxytocin treatment were applied during the third trimester of pregnancy, and females examined on day 6 and day 21 postpartum. The results indicated that serotonin levels were significantly increased on day 6 following unpredictable novelty stress during pregnancy. Furthermore, a marked reduction in serotonin levels in the hypothalamus, hippocampus, midbrain and brainstem was observed as a result of oxytocin treatment on both Day 6 and Day 21 postpartum. These results are in stark contrast to those obtained for nulliparous females in a previous study and suggest an important distinction between nulliparous and lactating females with respect to the effects of psychological stress and oxytocin treatment on central monoamine levels.

Emotional responsiveness is reduced in adult animals which have been exposed to stress during their first few weeks of life (1, 2). The stress of ‘handling’, daily removal of pups from their mother during the pre‐weaning period, also leads to a reduced corticosterone response to novel stimuli in adult life (3). In rats, exposure to novel stimuli results in the concomitant release of prolactin (PRL) and corticosterone (4–6). Here we show that, in male rats handled daily during the pre‐weaning period of life and tested in adult life for their hormonal responses to exposure to novel audio‐visual stimuli, the consequent secretion of adrenocorticotrophin (ACTH) is attenuated, but that of PRL is not. Thus, pre‐weaning handling results in permanent changes in a neural system specific to the control of ACTH secretion rather than affecting pathways common to neuroendocrine responses to emotional stimuli (7).
