Influence of inflammasome NLRP3, and IL1B and IL2 gene polymorphisms in periodontitis susceptibility

Abstract: The pathogenesis of periodontitis (PD) involves several molecules of the immune system that interact in a network to eliminate the periodontopathogens, yet, they contribute to periodontal tissue destruction. The different mechanisms that lead to periodontal tissue damage are not clear. Despite this, immune response genes have been related to the development of PD previously, such as those involved in inflammasomes which are multiprotein complexes and cytokines including Interleukin-1. The aim of the study was … Show more

“…IL16 polymorphisms were previously studied in PD [23,24]. Other SNPs in cytokine genes such as IL-1β, IL-2, IL-17 and IL-18 were associated with PD in our population from southern Brazil [25][26][27].…”

“…Negative inflammasome regulators, such as POP1, POP2, and CARD18, are also downregulated in periodontitis, further indicating increased inflammasome activity ( 225 ). Polymorphisms in various inflammasome components, including NLRP3, AIM2, and IFI16, may be associated with susceptibility to periodontitis ( 226 229 ). The mRNA levels of murine Ifi204 , which has a structure and function similar to those of human IFI16 , were also increased in gingival tissues of ligature-induced periodontitis ( 230 ).…”

“…1,6 Additionally, inflammasome polymorphisms were recently shown to put men at greater risk for developing periodontitis. 25 Further understanding of the role of the inflammasome in disease pathogenesis will help lead to precision care. Based on our knowledge thus far, individuals within the Socransky trait may benefit by adjunct therapeutic inhibition of the inflammasome to treat periodontitis.…”
