Influence of Low Molecular Weight Salts on the Viscosity of Aqueous-Buffer Bovine Serum Albumin Solutions

Abstract: Pharmaceutical design of protein formulations aims at maximum efficiency (protein concentration) and minimum viscosity. Therefore, it is important to know the nature of protein-protein interactions and their influence on viscosity. In this work, we investigated the dependence of the viscosity of BSA in an aqueous 20 mM acetate buffer at pH = 4.3 on protein concentration and on temperature (5–45 ∘C). The viscosity of the solution increased with protein concentration and was 230% higher than the viscosity of the… Show more

“…To answer the above two questions, we used a standard Nd:YAG laser with a pulse duration of approximately 10 ns. Bovine Serum Albumin (BSA) was used as a model protein, as it is one of the most studied proteins, quite frequently used in biological studies [ 23 ]. Thus, in this work, for the first time, using the example of BSA protein, it was discovered how protein molecules affect the characteristics of optical breakdown in an aqueous medium, how the optical properties of protein molecules change after optical breakdown, and what changes occur in the structure of molecules.…”

“…This involves taking into account both the properties of the involved fluids and the geometry of our system. The culture media solution has density ρ = 1008 kg/m 3 and viscosity η = 0.733 × 10 −3 Pa•s [41], and the aqueous 0.1 M acetate buffer has ρ = 1007 kg/m 3 and η = 1 × 10 −3 Pa•s [42]. The culture medium was modelled as solvent solution entering Inlet 1, the acetate buffer was modelled as dilute species entering Inlet 2 with a diffusion coefficient in the culture medium of 1 × 10 −9 m 2 /s.…”
