Influence of Strain Specificity and Culture Conditions on Terpene Production by Fungi

Abstract: The volatile oil production of several species and strains of the genus Ceratocystis (Ascomycetes) and of Lentinus lepideus (FR.) FR. (Basidiomycetes) is studied. The production of mono- and sesquiterpenes by these fungi proves to be highly influenced by strain specificity and culture conditions.

“…Pre-cultures of C. cinerea were prepared from a mycelium block (1 cm 2 ) as inoculum, which was excised from the agar plate and placed in a 300 mL Erlenmeyer flask containing 100 mL malt extract medium. After treatment with an Ultra Turrax homogenizer (Ika Werke GmbH & CO. KG, Staufen, Germany) the culture was incubated for 3 days at 150 rpm and 24 • C. For the production of lipolytic enzymes, malt extract medium, standard nutrient liquid (SNL) medium without glucose [24], and a mineral salt medium [25] were tested as main culture media to optimize the enzyme production. The main cultures were prepared in 500 mL Erlenmeyer flasks containing 250 mL medium, which was inoculated by transferring 20 mL of the pre-culture into each flask.…”
