Influence of thidiazuron (TDZ) pretreatment of shoot tips on shoot multiplication and ex vitro acclimatization of Harpagophytum procumbens

Abstract: The effects of thidiazuron (TDZ) pretreatment of shoot tips on Harpagophytum procumbens shoot proliferation and successive stages of micropropagation, i.e. rooting of regenerated shoots and acclimatization of plantlets to ex vitro conditions, were described in the present study. The best response in terms of shoot proliferation (about seven shoots/explant) and shoot length (3.2 ± 0.4 cm) was obtained when explants pretreated with 25 lmol L -1 TDZ for 6 h were cultured on Schenk and Hildebrandt medium containin… Show more

“…At the same concentration, BA induced 5.3 ± 0.26 shoots in 80% cultures, while Kin produced 3.6 ± 0.35 shoots with 71% regeneration frequency ( Table 1). The stimulating effect of TDZ on multiple shoot formation has been reported earlier for different medicinal plant species (Sivenesan et al 2011;Guo et al 2011;Grabkowska et al 2014;Singh and Dwivedi 2014). All regenerated shoots were free from any basal callusing.…”

“…TDZ-induced shoot organogenesis from different explants of many recalcitrant species as well as from woody plants has been reported earlier (Liu et al, 2004;Dewir et al, 2010Dewir et al, , 2011El-Mahrouk et al, 2010;Shaik et al, 2010). However, TDZ proved effective for in vitro propagation of many plant species (Huetteman and Preece, 1993;Baskaran and Jayabalan, 2009;Parveen and Shahzad, 2011;Grąbkowska et al, 2014) but not effective for others (Dewir et al, 2006b;Mitras et al, 2009) i.e. it is species specific.…”

“…Untuk tanaman berkayu kombinasi BA dan TDZ banyak digunakan dan memberikan hasil yang baik dalam memproduksi tunas in vitro. Grabkowska et al (2014) melaporkan bahwa TDZ dapat menginduksi pembentukan tunas adventif, proliferasi tunas adventif pada tanaman Harpagophytum procumbens.…”
