In the present study, due to the complex and numerous targets of Sarcandrae Herb (also known as Zhong Jie Feng), network pharmacology was performed to analyze its therapeutic effect on 2 cervical cancer cell lines, which could assist with the development of novel therapies. The results suggested that the natural flavonoid quercetin (Que), the effective antitumor ingredient in SH, which is widely present in a variety of plants, may depend on the target, EGFR. Previous studies have shown that EGFR serves a crucial role in the occurrence and development of cervical cancer, but its downstream molecules and regulatory mechanisms remain unknown. The anti-cervical cancer cell properties of Que, which are present in ubiquitous plants, were examined in vitro to identify the association between Que and its underlying pathway using MTT assays, flow cytometry, western blot analysis and Transwell assays. It was found that Que reduced cervical cancer cell viability, promoted G 2 /M phase cell cycle arrest and cell apoptosis, as well as inhibited cell migration and invasion. The Tyr1068 phosphorylation site of EGFR and the corresponding ERK target were also examined and the 2 kinases were markedly activated by Que. Furthermore, the EGFR inhibitor, afatinib and the ERK inhibitor, U0126 blocked the increase of EGFR and ERK phosphorylation, and resulted in a notable enhancement of apoptosis and cell cycle arrest. Therefore, to the best of our knowledge, the current results provided the first evidence that EGFR and ERK activation induced by Que could resist Que-induced anticancer activities. On this basis, the present study determined the role of EGFR and the underlying signaling pathways involved in the anti-cervical cancer malignant behavior induced by Que and identified the negative regulatory association.

Antiproliferative and Proapoptotic Effect of Daucus carota in Cervical Cancer Cells: An In Vitro Approach

Cervical cancer is the fourth most common cancer in women. Synthetic chemotherapeutic drugs such as bevacizumab have been employed for the treatment of cervical cancer, however, due to the associated side effects of bevacizumab, it has been substituted for plants with anticancer activities. Therefore, organic agricultural plants with anticancer activity have gained significant attention and emerged as a main area of research. Daucus carota (carrots) have phytochemicals that exhibit remarkable anticancer properties (antiproliferative and proapoptotic effects). We herein report the extraction of phytochemicals from D. carota. The characterization was performed using gas chromatography‐mass spectrometry. The inhibitory concentration at 50 % (IC50) of phytochemicals was assessed by sulforhodamine B assay utilizing HeLa cells and found 1.94 μg/mL. These cell lines were treated with varying concentration of phytochemicals (1.98 (1X IC50) and 3.88 μg/mL (2X IC50)) and were stained with 4′,6‐diamidino‐2‐phenylindole (DAPI) and propidium iodide (PI) dyes. Moreover, activation of caspase‐3 assay, changes in mitochondrial membrane potential, production of lactate dehydrogenase and reactive oxygen species, investigation of DNA damage by comet assay and cell cycle assessment by flow cytometry were accomplished. The results demonstrated that these phytochemicals led to apoptosis, cell shrinkage, chromatin condensation and nuclear fragmentation, triggered antiproliferative and proapoptotic potential on HeLa cell lines.

The term Traditional Chinese Medicine Constitution denotes the unwavering of the body's physical and psychological features. Unbalanced TCMCs generate vulnerability to diseases. This investigation examined the dissemination of the TCMCs of Chinese healthy women and those with breast cancer in Hong Kong to appraise the TCMC efficiency. Healthy woman (n=305) and women with BC (n=305) were recruited for this study to analyze the Physical Constitution Scale in TCM, thereby determining the types of TCMC. The sociodemographic data and connected BC risk factors were prepared using questionnaires. The association of unbalanced types of TCMC with BC was determined by multiple factor analysis. The percentage of unbalanced TCMC viz., Qi-depressed, Yin-deficiency, Blood stasis, Qi-deficiency, and Wetness-heat in BC patients was significantly greater than in healthy individuals (p<0.05). The analysis of stepwise logistic regression specified the constitution of Qi-depressed was positively correlated with BC (CI 1.49-6.92; OR=3.21). The correlation noteworthy enhanced when the Qi-depressed constitutions shared with Wetness-heat OR=4.82) or Blood-stasis OR=3.27). The grouping of both constitutions was connected to BC (CI 1.02-8.17; OR=2.88). The constitution of qi-depressed was an independent risk factor for BC. Similarly, the constitutions of Wetness-heat and Blood-stasis could also act as major functions in the incidence of BC when grouped with Qi-depressed. This outcome suggests that the unbalanced types of TCMC might be inconsistent in the individual with vulnerabilities to BC, proposing the possibility of Chinese constitutional analysis a potential strategy in detecting the vulnerable populace to BC and the corresponding inhibition of BC.
