Inhibition of Backpropagating Action Potentials in Mitral Cell Secondary Dendrites

Abstract: The mammalian olfactory bulb is a geometrically organized signal-processing array that utilizes lateral inhibitory circuits to transform spatially patterned inputs. A major part of the lateral circuitry consists of extensively radiating secondary dendrites of mitral cells. These dendrites are bidirectional cables: they convey granule cell inhibitory input to the mitral soma, and they conduct backpropagating action potentials that trigger glutamate release at dendrodendritic synapses. This study examined how mi… Show more

“…Olfactory bulb slices were prepared and visually identified cells recorded with techniques similar to those previously described (Lowe, 2002;Ma and Lowe, 2004). Briefly, male rats (P21-42, CD, Charles River Laboratories, Wilmington, MA) were sacrificed by decapitation while anaesthetized either by halothane inhalation, or by sodium pentobarbital (50-65 mg/kg, i.p.)…”

“…Slice preparation and recording followed previously described protocols (Lowe 2002). Briefly, male rats (P21-28, CD, Charles River Breeding Laboratories, Houston TX) were killed by halothane anesthesia, and the olfactory bulbs removed into an ice-cold sucrose slicing solution containing (in mM): 240 sucrose, 2.5 KCl, 10 Na-HEPES, 10 glucose, 1 CaCl 2 , 4 MgCl 2 , 0.2 ascorbic acid, pH 7.2, bubbled with 100% oxygen.…”
