Inhibition of Fam114A1 protects melanocytes from apoptosis through higher RACK1 expression

Abstract: Fam114A1 is a gene closely related to the development of nerve cells, melanocytes, and nerve cells that originate from the neural crest of the embryonic ectoderm. Recent studies showed that Fam114A1 has a role in the occurrence of ankylosing myelitis spondylitis and autoimmune enteritis; still, its cellular function remains poorly understood. In this study, we investigated the effect of Fam114A1 on the biological activity of melanocytes. We found that the expression of Fam114A1 in vitiligo melanocytes (MCV-L, … Show more

“…Apoptosis is a type of cell death that depends on genetically coded signals or activity within dying cells. It is concerned with the growth and development of organs and tissues, immunity, metabolism and the removal of abnormal cells [ 34 , 35 ]. Furthermore, we observed that more cellular debris at the bottom of the medium, and “drawing” of the cells morphologically under the microscope after MH treatment, and the number of AML12 cells was reduced in a time-dependent manner after MH treatment.…”

“…When we explored previous studies on FAM114A1 to understand its function, particularly under hypoxia, we found that apart from its role in neuronal cell development [ 13 ], melanocyte apoptosis [ 32 ] and an association with ankylosing spondylitis [ 33 ], there was very little known about this gene or its homologue FAM114A2 . A recent study in Fam114a1 −/− mice showed that cardiac function was markedly restored in the knockout mice, in angiotensin II infusion model of hypertension, when compared to the controls [ 29 ], indicating its role in oxidative stress.…”

“…RACK1 is a highly conserved tryptophan-aspartate repeat (WD-repeat) protein and is currently recognized as a multifunctional scaffold protein that is involved in a wide range of biological processes, such as viral infection [ 17 ], cell migration [ 18 ], and apoptosis [ 19 ]. It has been reported that RACK1 participates in NLRP3 inflammasome activation by promoting NLRP3 conformational changes and inflammasome assembly [ 20 ].…”
