Inhibition of Hec1 expression enhances the sensitivity of human ovarian cancer cells to paclitaxel

Abstract: Aim: Hec1, a member of the Ndc80 kinetochore complex, is highly expressed in cancers. The aim of this study was to explore the role and mechanism of action of Hec1 with respect to the cytotoxicity of paclitaxel in ovarian cancer. Methods: Thirty ovarian cancer samples and 6 normal ovarian samples were collected. Hec1 expression in these samples was determined with immunohistochemistry. Ovarian cancer cell lines A2780, OV2008, C13K, SKOV3, and CAOV3 and A2780/Taxol were examined. Cell apoptosis and cell cycle a… Show more

“…Expression of the mitotic regulator NDC80 is increased in various human malignancies, including HCC. Considering the significant overexpression of NDC80 in multiple human malignancies[ 23 - 27 ], studies on the mechanisms of NDC80 overexpression are significant. Liu et al[ 28 ] also reported that the expression level of NDC80 was remarkably up-regulated in HBV-related HCC tissues.…”

“…Even though the site of action is very closely associated with microtubules, the potency profile shows that the mechanism of action of TAI-95 has interesting features that may give it an advantage over conventional cytotoxic drugs. A study has showed that Hec1 overexpression was associated with paclitaxel resistance and poor prognosis in ovarian cancer (28), further highlighting the role of the target, Hec1, in drug-resistant cancers. Even though more studies will be necessary to elucidate the underlying mechanisms, the potential of this drug draws immense interest and we hold great expectations on its impact on the clinical treatment of drug-resistant breast cancers.…”
