Background Zinc finger and BTB domain-containing 7A (ZBTB7A) is a member of the POK family of transcription factors that plays an oncogenic or tumor-suppressive role in different cancers depending on the type and genetic context of cancer. However, the function and molecular mechanism of ZBTB7A in bladder cancer (BC) remain elusive. Methods The role of ZBTB7A in bladder cancer was detected by colony formation, transwell, and tumor formation assays. The expression levels of ZBTB7A, HIC1, and miR-144-3p were analyzed by qRT-PCR and Western blot. Bioinformatics analysis and a dual-luciferase reporter assay were used to assess the effect of ZBTB7A on the promoter activity of HIC1. Results The present study revealed that knockdown of ZBTB7A suppressed BC cell growth and migration, as indicated by an approximately 50% reduction in the number of colonies and an approximately 70% reduction in the number of migrated cells. Loss of ZBTB7A inhibited tumor growth in vivo, resulting in a 75% decrease in tumor volume and an 80% decrease in tumor weight. Further mechanistic studies revealed that ZBTB7A bound to the hypermethylated in cancer 1 (HIC1) promoter and downregulated HIC1 expression, accelerating the malignant behavior of BC. Increased expression of ZBTB7A in BC tissues was negatively corrected with the expression of HIC1. Moreover, ZBTB7A was a target of miR-144-3p, which decreased ZBTB7A expression in BC. Conclusion Our data demonstrate that ZBTB7A, a targeted gene of miR-144-3p, promoted tumorigenesis of BC through downregulating HIC1 expression.

With the widespread use of highly active antiretroviral therapy (HARRT), the survival time of AIDS patients has been greatly extended. However, the incidence of lung cancer in HIV‐infected patients is increasing and has become a major problem threatening the survival of AIDS patients. The aim of this study is to use Weighted Gene Co‐expression Network Analysis (WGCNA) and differential gene analysis to find possible key genes involved in HIV‐infected lung cancer. In this study, using lung tissue samples from five pairs of HIV‐infected lung cancer patients, second‐generation sequencing was performed and transcriptomic data were obtained. A total of 132 HIV‐infected lung cancer‐related genes were screened out by WGCNA and differential gene expression analysis methods. Based on gene annotation analysis, these genes were mainly enriched in mitosis‐related functions and pathways. In addition, in protein–protein interaction (PPI) analysis, a total of 39 hub genes were identified. Among them, five genes (ASPM, CDCA8, CENPF, CEP55, and PLK1) were present in both three hub gene lists (intersection gene, DEGs, and WCGNA module) suggesting that these five genes may become key genes involved in HIV‐infected lung cancer.

Liquid biopsy has advantages over traditional biopsy, which cannot determine tumor dynamics. As a noninvasive and precise test, liquid biopsy detects biomarkers that carry information on tumor progression and has undergone tremendous development in recent years. Exosome detection is one of the methods of liquid biopsy. Radiotherapy affects the release of exosomes and intercellular communication. Based on the properties, extractability, and detectability of exosomes, key exosomal cargoes after tumor radiotherapy can be used as biomarkers for tumor prognosis. Exosomes after tumor radiotherapy can be used for liquid biopsy. The main applications include (1) predicting radiotherapy efficacy, (2) predicting tumor prognosis, and (3) optimizing the regimen of tumor treatment. This review provides further research directions for liquid biopsy after tumor radiotherapy.
