Inhibition of Neuroinflammation by AIBP: Spinal Effects upon Facilitated Pain States

Abstract: SUMMARY Apolipoprotein A-I binding protein (AIBP) reduces lipid raft abundance by augmenting removal of excess of cholesterol from the plasma membrane. Here, we report that AIBP prevents and reverses processes associated with neuroinflammatory-mediated spinal nociceptive processing. The mechanism involves AIBP binding to Toll-like-receptor-4 (TLR4) and increased binding of AIBP to activated microglia, which mediates selective regulation of lipid rafts in inflammatory cells. AIBP-mediated lipid raft reductions … Show more

“…These cell culture experiments were corroborated with experiments in embryonic zebrafish in which apoa1bp knockdown resulted in ectopic and excessive angiogenesis (22). Similar to the effect of AIBP on endothelial cells, AIBP has been reported to promote cholesterol efflux from THP-1derived and RAW macrophages and microglia (23)(24)(25).…”

“…Overexpression of AIBP delivered by an adeno-associated virus reduces atherosclerosis in hypercholesterolemic mouse models (24,39). In addition, spinal delivery of recombinant AIBP prevents neuropathic pain states in mice (25).…”

“…54,55 This is in line with the data that TLR-4 interacts with ApoA-I binding protein, recently shown in a pain model. 11 Oral D-4F is safe and well tolerated by high-risk cardiovascular patients. The daily application of single doses of D-4F reduced their HDL-inflammatory index.…”

“…Most amyloidogenic proteins concentrate and sometimes processed in lipid rafts, initiating the rollover of neurodegeneration (10). Raft mitigation strategy using βcyclodextrins and Apolipoprotein A-I Binding Protein (AIBP) were successfully tested in restricting inflammation, neuropathic pain, atherosclerosis and HIV infection (11)(12)(13)(14), but delicate balance must be preserved not to affect physiological pathways relying on lipid rafts.…”
